Object: To explore the expression of immunoglobulin E(IgE), interleukin-5(IL-5) and.eosinophil in nasal polyp and their roles in the pathogenesis of nasal polyp.Method: 38 patients with nasal polyp and 15 control cases were chosen. Their IL-5 concentration in tissue homogenate were measured by enzyme-linked immunosorbent assay (ELISA). All samples were stained with Chromotrope 2R histochemical and SP immunohistochemical method separately to examine eosinophil and IgE+ cells in nasal polyp.Result: (1) The level of IgE, IL-5 and eosinophil in nasal polyp was all significantly higher than that in normal control specimens (P<0.01). (2) The positive correlation of IgE and eosinophil was significant (r=0. 843, P<0.01). And the positive correlation of IL-5 and eosinophil was significant too (r =0.642, P < 0.01). At last the positive correlation of IL-5 and IgE was significant (r =0.667, P < 0.01).Conclusion: Nasal polyp is a disease characterized by eosinphils. IgE and IL-5 which assemble and activate eosinophils play an important role in the pathogenesis of nasal polyp.
