Obejective: To observe the effects of the acupuncture on the concentrations of endothelin and Nitric Oxide in the plasma of migraine rats, and explore their relativity. To get the further exploration of the mechanism of the acupuncture in treating migraine.Method: Sprague-Dawlay rats(n=40) were divided ramdomly into four groups: â‘ Normal control group(n=10); â‘¡Model control group(model was reproduced by electrical stimulation of trigeminal ganglion, not treated with acupuncture,n=10); â‘¢ Acupuncture control group (model reproduced ,then treated with acupuncturing non-channel points, n=10); â‘£Acupuncture treatment group (model reproduced,then treated with acupuncturing Taichong and Yanglingquan points, n=10).The blood from external jugular veins(EJV) was taken after the experiments had finished, the contents of plasma endothelin(ET) and Nitric Oxide(NO) were differently determined with radioimmunoassay(RIA) method or reductase method, and the relativity of them was analyzed.Result: â‘ The contents of plasma ET and NO in each group: The ET and NO contents of the blood in EJV were constantly kept lower level in normal rats; Compared with normal control group, the levels of ET and NO in model control group were significantly increased( P<0.01); Compared with model control group and acupuncture control group, the levels of ET and NO in acupuncture treatment group were significantly decreased (P<0.01); No significant difference was found betweenacupuncture control group and model control group(P>0.05). â‘¡It was showed that the plasma levels of ET have a close positive correlation with the plasma levels of NO during headache attack periods (r=0.663, P<0.05).â‘¢ ET/NO ratio of each group: ET/NO ratio of model control group was significantly decreased than that of normal control group(P<0.01); Compared with model control group and acupuncture control group, ET/NO ratio of acupuncture treatment group was both significantly increased (P<0.01); No significant difference was found between acupuncture control group and model control group(P>0.05).Conclusion: â‘ According to Knyihar-Csillik way the experimental migraine rats model was reproduced successfully by electrical stimulation of trigeminal ganglion. â‘¡It was showed that experimental migraine rats might have a increased release of ET and NO, it was observed that ET/NO ratio was decreased, the attack of migraine was accounted for the dominant biology action and functional intensity of NO. The unbalance between vasoconstrictor neuropeptide ET and vasodilatatior neuro-transmitter NO might play an important role in the pathogenesis of migraine. â‘¢ It was suggested that the needling sensation by the stimulation of acusector could act reflectly neurol all levels, remove and excitate the latent self-regulate mechanism of organism via by the relation between points and The Channels and Collaterals, reduce the contents of ET and NO, adjust ET/NO ratio in the plasma of migraine rats model, it may be one of the mechanisms in treating migraine with acupuncture.
