Objective: Transient forebrain ischemia produced by four-vessel occlusion (4-VO) triggers the delayed death of CA1 neurons in the hippocampus, resulting in deficits of learning and memory. In this study, the purpose is to investigate effect of YuShenKouFuYe(YSKFY), a traditional Chinese medicinal prescription, on delayed neuronal death of hippocampus and learning and memory deficits after transient global forebrain ischemia in rats. Methods: Transient forebrain ischemia was produced using published modifications of the four-vessel occlusion (4-VO) method. YSKFY was given orally to rats. The learning and memory was tested through finding a hidden clear plastic platform that was placed 33 cm away from the wall of the water maze and 1 cm below the water. Then, we studied the effect of YSKFY on cell survival in the hippocampus and learning and memory in a rat model of transient global ischemia.Results:1, Oral administration of YSKFY has effect on cell survival. However, YSKFY significantly intensified the number of survival neurons.2 , YSKFY significantly attenuates ischemia-induced learning and memory deficits. Conclusion: The present results suggested that YSKFY given orally prevented the delayed neuronal death of hippocampus and improved the learning and memory of transient global forebrain ischemia in rats.
