With the development of the aged population, senescence is more and more importance and big problems to the Chinese. To find out the essential of senescence and high power anti- senescence drugs are became front in the geratology field.This article was discussed the senile changes of the neuron, the effects of Cnidium offcinal Makino and potential mechanisms of senescence by using senile neuron caused by D-galactose in-Vitro cultured rat cerebroneuron. Giving the basic data to anti-senescence study, the value of the Cnidium offcinale Makino on the therapy and develop the Chinese traditional medicine for future.The experiment was set up 6 groups: Normal control group; D-galactose group; treated by Cnidium offcinale Makino group was subdivided into large, middle, small dose group; Renshenzaidai group. Cultured the neuron for 10 days, the growth of the cultured cells were observed by inversion microscopy; the changes of the ultra structure were observed by electronic microscopy; the changes of the Nissl bodies and LPF contents and the activation of SDH and ACPase on cultured cells bycytochemical detection technology. The results: D-galactose can significantly inhibit cultured neurons and increase the mortality of the neurons, destroy the ultra-structure, decrease the Nissl bodies' contents, increased LPF deposit, decreased the activation of the SDH(P<0.05), increased the ACPase activation(P<0.01). The difference dose of the Cnidium offcinale makio had no significant difference.The results of the experiment showed: D-galatose can increased the aging of the cultured rat neurons; Cnidium offcienale Makino have the defended effects on rat cultured neurons in vitro; Cnidium offcienale Makino can do good effects on the senile neurons caused by D-galatose.
