OBJECTIVE: To study the protective effect of Ligusticum acutiloba, Cnidium offcinale, Gentiana manshunca, Bupleurum Chinese, Rhodiola dachaliensis on acute liver injury in mice. METHODS: acute liver injury models induced by D-galactosamine(D-GalN) /Lipopolysaccharid(LPS) in mice, and the levels of serum enzyme ALT, AST on acute liver injury with pretreatment the aqueous and ethanol extract of Gentiana manshunca and Bupleurum Chinese, Rhodiola dachaliensisA, the ethanol extract of Ligusticum acutiloba, the aqueous extract of Cnidium offcinale were observed and we also observed the changes of liver histopathology RESULTS: Pretreatment of mice with the aqueous and etanol extract of Rhodiola dachaliensis, the ethanol extract of Ligusticum acutiloba, the aqueous extract of Cnidium offcinale, Gentiana manshurica, Bupleurum chinese( 100mg/Kg ,intraperittoneal) significantly reduced the levels of serum enzyme ALT, AST on acute liver injury', and have significant difference compared with control group ( P<0.001) .Althought the aqueous extract of Ligusticum acutiloba, the ethanol extract of Gentiana manshurica, Bupleurum chinese can reduce the levels of serum enzyme ALT. AST on acute liver injury, but the levels of serum enzyme ALT, AST have no significant difference compared with control group (P>0.05) . Histopathologic examination shows that the aqueous and etanol extract of Rhodiola dachaliensis, the ethanol extract of Ligusticum acutiloba, the aqueous extract of Cnidium offcinale, Gentiana manshunca, Bupleurum chinese can relieve liver injury in mice induced by Galn/LPS. CONCLUSION: the aqueous and etanol extract of Rhodiola dachal iensis, the ethanol extract of Ligusticum acutiloba, the aqueous extract of Cnidium offcinale, Gentiana manshunca, Bupleurum Chinese has protective effects for mice acute liver injury induced by D-Galactosamine and lipopolysaccharide.
