Objectives: Alzheimer's disease (AD),a progressive degenerative disease of the central nervous system, characterized by progressive memory and acquired knowledge loss. The characteristic change of pathology in AD were neuritic plaques(NP),neurofibrillary tangles(NFTs) and the loss of cholinergic neuron. In the present study, a method of stereotaxic apparatus was employed to damage the nucleus basalis of Meynert. Then we measured the behavior changes by Y maze and the changes of neurotransmitters in hippocampus after administration of prescription of regulating the heart and invigorating the kidney, and discussed the treatment of AD and their anti-dementia mechanism, providing a foundation of treatment on AD with Chinese medicine.Methods: l.Subjects:Wistar rats weighed from 300-350g were used, male only. 2.Building of AD models: Experimental animals were anesthetized by pentobarbital (30mg/kg, i.p.), then were placed on the brain stereotaxic apparatus and received a bilateral lesion of Meynert by LA injection, according to the atlas of Paxinos and Watson. After seven days of lesion, selected AD rats. 3.Y maze test: A quiet and dark room was needed in this experiment. Rats were tested in a trisecting, Y type maze. After the adaptation of three minutes in the maze, stimulations were given by clockwise with lamplight whose voltage was 30-60V (lamplight represent safe area, the other two were stimulus area). When the rats escaped to the safe area, lamplight was still in work for another 15 second, then the light stopped and 1 minute's rest. After that, the second stimulation began. The stimulation was repeated 40 times in order, times of right and wrong were recorded. Memory ability was tasted 48 hours after learning test with the same methods. 4.Measurement of neurotransmitters: Rats were decollated, then extracted hippocampus, and placed in liquid nitrogen. All the aboveABSTRACToperations should be done as rapidly as possible. Serotonin and Norepinephrine were measured by high performance liquid chromatography (HPLC), and Acetylcholine by spectrophotography techniques.S.Statistical analyses: All the data were done by methods of t-test and single factor analysis of variance with SPSS 9.0 statistical procedure. Significance level was chosen at 0.05 and all tests were two-sided.Results: 1. After the injection of LA into nucleus basalis of Meynert, the learning and memory scores decreased, there were significant difference ( P <0.01,P<0.01) between model group and model control group; The levels of Acetylcholine, Serotonin, Norepinephrine in hippocampus decreased comparing to model control group, there were significant difference (P<0.01,P<0.01). 2. After 20-day' s gavage, the learning and memory scores of dementia rats increased. Learning scores were 1.57 times higher than before and memory scores were 1.54 times higher than before, there were significant difference(F<0.01^P<0.01 ) .3. After 20-day' s gavage,the learning and memory scores of dementia rats increased. Learning scores were 1 .40 times higher than before and memory scores were 1.29 times higher than before, there were significant difference (KO. 01, KO. 01) . 4. After 20-day' s gavage, the learning and memory scores of dementia rats increased. Learning scores were 1.64 times higher than before and memory scores were 1.60 times higher than before, there were significant difference (/XO. 01, P\0. 01 ) .5. Prescription of regulating the heart and invigorating the kidney was super than single use on the treatment of AD, there were significant difference (/KO. 05, /KO. 05) , prescription of regulating the heart was super than that of invigorating the kidney, there were significant difference( KO. 05, KO. 01 ) 6.After the single and combining use of the prescriptions of regulating the heart and invigorating the kidney, the level of Acetylcholine, Serotonin, Norepinephrine increased compared with model group, there were significant difference (P<0.01).ABSTRACTConclusions: 1.
