Objects: Identification the prognosis of patients with glioma is important for selecting and evaluating the effectiveness of treatment. The aim of this study was to evaluate the Topo II a labeling index(LI) using the Topo II a monoclonal antibody(MOAb) as a prognostic indictors for patients with glioma and establishing a correlation with clinicopatbological parametersand survival. Method and Materials: Topo II a and PCNA immunohistochemically were performed on paraffin secions to estimate the growth potential of 41 gliomas as using MoAb. Calculated the labeling index(L1) for Topo II a and PCNA. The finding were correlated with clinopathological features of the patients ,including their clinical outcome. Results: The proliferative labeling indices based on cells staining for PCNA and Topo ~I a correlated highly with each other(r=0.981,p<0.O1). The TopoII a LI was:l1.39?.38 in grade2 glioma group(n11), 38.83?10.9lin the grade3 group(n=19), and 47.59 ?10.13 in the grade4 group(n~=1 1), representing significant difference between low grade gliomas and grade3,grade4 gliomas(p0.05). Topo II a LI more than 40% III L~t ~b~fl~1I a predicted shorter survival, but Topo II a LII less than 10% predicted longer survival. Conclusion: These results indicate Topo II a LI obtained using topoisonierase II a MoAb is an important and useful tool for predicting survival.
