ObjectiveThe purpose of this study was to examine the expression of PCNA and MMP - 9 in human primary and recurrent glioma. investigate the relationships among expression of them and glioma histopathologic grades and recurrence of gliomTis-sues.Methods1. Patient selectionMatched - pairs tissue samples of primary and recurrent Glioma ( n = 38 ) were obtained from patients who underwent surgery in the affiliated hospital of China Medical University between 1998 and 2003. Non - tumor brain tissues ( n = 10) were obtained from patients who underwent surgery in the Second affiliated hospital of China Medical University between January, 2004 and March, 2004. All tumors were graded according to World Health Organization criteria (2000).2. ImmunohistochemistryA total of 48 formalin - fixed and paraffinembedded specimens was available for this study. Expression of PCNA and MMP - 9 was immuno - Histochemi-cally investigated, namely, the streptococcus antibiotic protein -peroxidase(S -P process) method.3. Result determinationIf brown chromatingranule appears in the nucleus,the chromaticity is positive for PCNA, brownish staining of cytoplasm,vascular basement membrane andendothelial cells is regarded as MMP -9 positive. Under the fluorescope, randomly take five high - powered fields of vision to count the positive cell percentage of PCNA and MMP -9. 4. Statistical methodsThe analysis is progressed with statistics software SPSS. Independent samples t test was used to compare the expression of PCNA and MMP - 9 in different pathology garde. Paired - samples t test was used to compare the expression of PCNA and MMP - 9 in primary and recurrent glioma. Wilcoxon signed rank test was used to compare the pathology garde of primary and recurrent glioma. Non - parametric Spearman rank correlation coefficients was used to compare the expression of PCNA and MMP - 9 to pathology garde, assess the degree of association between PCNA and MMP -9. Satistical significance defined as p≤0. 05.ResultsTissue samples of non - tumor brain ( n = 10) were negative for both PCNA and MMP -9. PCNA was expressed except individual case,MMP -9 was positive in all of the 38 glioma tissue samples. The expression of PCNA has a significant difference between singulorum pathology garde (p <0.05). The expression of MMP - 9 has a significant difference between grade 1, grade 2 and grade 3 glioma, but its not significant difference between grade 3 and grade 4 glioma ( p >0.05) . We also analyzed the relationships between the histopathologic grades of glioma, expression intensity of PCNA and MMP -9. The PCNA and MMP -9 were also trend increase with glioma grade (p <0.01) , but there was no correlation between PCNA and MMP - 9 (r = 0.227 p > 0.05 ).There have 4 grade 1, 7 grade 2, 8 grade 3 in primary glioma. There have 12 increased, 4 fixed and 3 induced in the histopathologic grades of the recurrent glioma (p <0.05). Glioma recurrent time has a correlation with PCNA labelling index, it means PCNA LI more high, glioma recurrence is more fast.Use Paired - samples t test to compare the expression of PCNA and MMP -9 in primary and recurrent glioma, found they also tended to increase with tumor s recurrence (P_pcna <0.05, Pmmp-9 <0.01).DiscussionPCNA is an auxiliary protein of DNA pclymerase δ . Expression intensity of PCNA is closely related to tumor cell proliferation, it' s at equal pace with DNA synthesis. MMP -9 is one of the matrix metalloproteinases, impersonate it' s region invasion in glioma biological behaviour, and its related to tumor angiogene-sis.In this experiment, PCNA was expressed except non - tumor tissue and individual case, its expression intensity is trend increase with glioma grade. MMP -9 was positive in all of the glioma tissue samples. PCNA and MMP -9 were also trend increase with glioma grade. We also analyzed the relationships between the expression intensity of PCNA and MMP - 9, display there was no correlation between PCNA and MMP - 9 ( r = 0. 277, P > 0. 05 ). It explain tumor cell multiplication and invasion has different occurrence mechanism, or they betide in different cycle, materialize different bionomics of malignancy.Wilcoxon signed rank test was used to compare the pathology garde of primary and recurrent glioma ( p <0.05) , use paired - samples t test to compare the expression of PCNA and MMP - 9 in primary and recurrent glioma ( P_pcna < 0.05, Pmmp 9 <0.01) , also possess statistical significance.
