Objective:1.The significance of HPV and TCT in screening cervical lesions was preliminarily analyzed through the statistics of HPV and TCT report results in each group.2.By testing whether there was positive expression and the expression level of ToPo? alpha in each group,as well as the expression of P16 and Ki-67 in each group,to investigate the relationship between the expression levels of the three markers and diffent levels of cervical lesions.3.To analyze the value of the three molecular markers alone or in combination in the early diagnosis and grading diagnosis of cervical precancerous lesions.Methods: Cervical tissues with cervical lesions admitted to handan central hospital from January 2018 to January 2019,were collected retrospectively with additionally,analyzed the HPV types and TCT report results by statistical method,then the results of routine HE pathology were verified again by two pathologists,divided into CIN? group46 cases,CIN? group 33 cases,CIN? group34 cases based on the severity of cervical lesions.At the same time,45 cases of normal cervical tissue were randomly selected as the control group.The positive expression of P16?Ki-67 and ToPo? alpha in different degrees of cervical lesion tieeue were detected by adopting the ImmunoHistochemietry method,to evaluate the significance of the three molecular markers mentioned above in the diagnosis and grading of cervical precancerous lesions.SPSS21.0 was used for data collation and analysis,and P < 0.05 showed a statistical difference.Results:1.The detection rate of high-risk HPV was 0.00% in the control group,higher in the cervical lesions,but there was no significant difference between the three cervical lesion groups(P>0.05).2.The positive detection rate of TCT was 0.00% in the control group,higher in the cervical lesion tissue,but there was no significant difference between the three cervical lesion groups(P>0.05).3.The positive expression rates of P16 in the control group,CIN? group,CIN?group,CIN? were 0.00%?34.78%?60.61%?85.29%,the positive expression rates ofki-67 in the control group,CIN? group,CIN? group,CIN? were 15.56%?45.65%?69.70%?94.12%,the differences between these four groups were statistically significant(P<0.05);the positive expression rates of ToPo ? alpha in the control group,CIN?group,CIN? group,CIN? were 11.11%?21.74 %?54.55 %?88.24 %,the difference was no statistically significant difference between the control group and CIN? group;whereas,the difference was significant in either the comparison of CIN?,CIN? and CIN? group(P< 0.05).4.The higher positive expression rate of P16 and Ki-67,ToPo ? alpha has significant correlation with the higher level of cervical lesion,the Spearman correlation coefficients were 0.678,0.670 and 0.647.5.The receiver operating characteristic curve results showed that the area under the curve of Ki-67 single detection is 0.792,higher than that of P16 single detection area under the curve and ToPo ? alpha separate inspection area under the curve,but there was no statistically significant difference between comparison(P>0.05),but the area under the curve of P16,Ki-67 and ToPo ? alpha combined detection was 0.845,higher than that of single detection of these 3 indexes,the difference was statistically significant(P<0.05).Conclusions: HPV and TCT could be preliminarily screened out cervical lesions,but but there was no advantage to distinguish the level of cervical lesions.As the same as P16 and Ki-67,there was a positively correlation between the positive expression of them and the severity of cervical lesions.The combined detection of the P16 and Ki-67,ToPo? alpha could improve the effect of the diagnosis of cervical lesions,and could be used for the grading diagnosis of cervical lesions and the prediction of cervical lesions.
