Neural cell adhesion molecule(NCAM) belongs to immunoglobulin superfamily,it located on cell membrane and acted as a kind of adhesion mediater to mediate intercellular adhesion of neuronal or glia cells in central nerve systeme(CNS) through adhesive to other molecules or to heparin or heparin sulfate.The relationship of the expression of NCAM and the invasiveness biological behavior was payed close attention.But the correlation of the expression of NCAM and the prognosis of the patients has not been found in the literature. Proliferative cell nuclear antigen (PCNA) was a kind of acid nucleoprotein which was the coenzyme of DNA polymerase and was essential during the synthesis of DNA in tumor cells.There was positive correlation between the expression level of PCNA and the pathological grade of glioma. To reveal whether there was any correlation between the proliferation activity and invasiveness of glioma the expression of NCAM,PCNA in 48 cases of glioma patients were detected. Further more, combined with clinical follow-up the feasibility of predicting glioma prognosis by NCAM,PCNA was discussed 6 Material and methods 1. Material The 48 specimens of glioma which were undergone operation during April 1996 to June 2000 in the first affliated hospital of Medical college Zhe Jiang university were examined by using immunohistochemical staining.There were 29 male and 19 femal with an age range from 11 to 71 years old and mean age was 40.73 years old.These specimens were classified by 2 pathologists as kernohan classification, and the results were grade I 4 cases,grade II 22 cases, grade III 15 cases and grade IV 7 cases. All the 48 cases were followed up until Mar. 2000. The follow-up time was range from 4 to 36 month with the mean time of l0.l1梞onth and 5 cases were lost. 2.lmmunohistochemical staining 2 step way of Dako-Envision immunohistochemistry was taken lust as manua. 6 cases normal brain tissue which were derived from internal decompression of cranial injury patients were used as positive control. Buffer instead of first and second antigen were used as negative control.Criteria:These cells whose cytoplasm or cell membrain stained brown-yellow or yellow were regard as positive in NCAM staining.While the cells with brown-yellow stained nuclear were regard as positive in PCNA staining.l0 high power fields were selected randomly and 1000 cells were counted then the percentage of positive cells was calculated named as NCAM-Pl or PCNA-PI respectively. 3.Statistic analysis The correlation of NCAM-Pl,PONA-PI and the pathological grade were analysed by Anova analysis.The correlation of NCAM-PI,PCNA-PI and half year survival rate were analysed by Chi-square test. The correlation of NCAM- PI,PCNA-PI and pathological grade were analysed by linear regression analysis. 7 Results The NCAM-PI was 61.67?2.29 in normal brain, the positive rate was l00%.The NCAM-PI was 45:00?3.80, 39.77?13.93 in grade I ,grade II glioma respectively and the positive rates were 100% both in grade I and grade II .The positive rate of grade III was 80.00% (12/15) with the NCAM-PL was 23.67?3.34.The positive r...
