Study On The Effect Of Different Soil Types On Soil Biological Activity Of The Rhizosphere And Non-rhizosphere In The Flue-cured Tobacco Growth

This article based on studies of yellow brown soil , alluvial soil, brown soil, lime concretion black soil and yellow cinnamon soil .After transplanting flue-cured tobacco six period, the field collected rhizosphere and non-rhizosphere soil, studied Rhizosphere and non-rhizosphere the number of microorganisms, soil enzyme activity, soil moisture content and cured tobacco leaf.Use molecular method(DGGE)to research the change of microorganism in the flue-cured tobacco of overluxuriant growth stage. The main conclusions are as follows:(1)General speaking, difference of microorganism quantity among different rhizosphere and non—rhizosphere types of soil is significant,flue-cured tobacco rhizosphere and non-rhizosphere micro-organisms have shown the number of the same law: bacteria> actinomycetes> fungi.The bacteria,fungi and actinomycetes total amount in rhizosphere and non—rhizosphere yellow brown soil were highest. The bacteria and actinomycetes total amount in rhizosphere yellow cinnamon soil were least. The potassium bacteria,aerobic azotobacter and ammonifier total amount in rhizosphere brown soil were highest. The phosphorus bacteria and bacillus total amount in rhizosphere yellow brown soil were highest.(2) Catalase activity and Urease activity have a single peak curve pattern with Maximum that occurred in different time in rhizosphere and non—rhizosphere soil. Invertase activity changed with a inclined inverted"V"mono peak variation in Rhizosphere Soil. The maximum values occurred in 40-60d after transplanting in rhizosphere and non—rhizosphere soil. General speaking, The Catalase activity and Invertase activity in rhizosphere yellow brown soil were highest. The Urease activity in rhizosphere lime concretion black soil were highest.(3) The contents of available nitrogen,available P,available K and organic matter was also wave-like in the soil moisture content with Maximum that occurred in 60-100d after transplanting.The soil moisture content have shown for the rhizosphere>non-rhizosphere ,rhizosphere significant effect(R/S).(4) Correlation analysis indicated that there existed a close relationship between the in Rhizosphere Soil of microorganisms,soil moisture content and soil enzyme activity. The soil1 microorgnaisms and soil moisture had a signifieani or highly signifienatre lationship with pH,hydrolyzable nitrogen,available phosphorous ,rapid available potassium and Organic Matter.The soil1 microorgnaisms had a significantly or very significantly correlated with Catalase, Urease and Invertase.(5) The filling value, thickness of leaves, dragging force, tensile strength were relatively suitable in different soil types.The ratio of total nitrogen to nicotine and ratio of total sugar to nicotine always were in optimum range in brown soil,the ratio of potassium to chlorine was kept with higher level in yellow brown soil. Aroma content of different soil types yellow brown soil > alluvial soil> brown soil > lime concretion black soil > yellow cinnamon soil.(6) Judging from the result of DGGE ,brown soil and yellow brown soil microorganism total quantity are bigger ,the bandings are many and their superiority population brightness are bigger than the other soil types. DGGE could clearly separatethe amplified fragments of 16S rDNA genes (V3 region)of different microbial community in soils,which make it possible to identify and define these 16SrDNA fragments by sequencing.
