Pleurotus eryngii is a kind of rare edible fungi. In order to breed strain with early maturity and high quality, we investigated the inheritable characters of twelve Pleurotus eryngii strains by performing mycelium growth rate test, resistance test and agronomic traits comparation and chose the strains with premature properties. high quality and yield as the parents. Single-core mycelium groups were built with the methods of single spore dilution separation, single-core mycelium mating response and ISSR marker analysis.and new hybrid strains were obtained through single×single hybridization. The high quality of the hybrid strain was verified by the mycelium growth rate test, resistance test. ISSR molecular marker, fruiting tests. The major results were listed as the following:By comparing the mycelia growth rate, growth potential, resistance, planting cycles, yield, and quality among the 12 strains of Pleurotus eryngii. it was found that mycelium of the Pe6 and the Pe7 grew faster; the Pe6, Pe7, Pe10, Pe11 exhibited strong resistance:The Pe3. Pe9. Pe10 had short planting cycle; The Pe11; Pe7. Pe9. Pe10 showed higher yield and the Pe2. Pe6. Pe7, Pe8, Pe12 had better quality and morphological appearance.The clustering analysis of the 12 Pleurotus eryngii strains was carried through by using ISSR molecular markers. The strains were divided into three groups when the coefficient of similarity was 0.83 and the Pe3 and Pe7 was chosen as parents through comprehensive analysis.By microscopic observation. antagonistic test and ISSR marker analysis. single-core mycelium was classified into eight different mating types derived from the two parents including two categories, one was the Pe3 for single-core mycelium of T1. T2. T3. T4 and the other was the Pe7 for single single-core mycelium of T5. T6. T7. T8.Twelve hybrids were obtained from single×single mode of eight kinds of mating types of single-core mycelium by microscopying. complementary bands and new hybrid bands on electrophoresis image.Comparative test of mycelium, resistance test and fruiting tests showed that T4T7 and T2T7 were ideal hvbrid combinations.
