This paper studied the polyphenols'extraction from peanut skin and compared the polyphenols'difference between 8 kinds of peanut skin with the methods of FC and HPLC. At the same time, this paper studied the peanut skin polyphenols efficacy of anti-AA to mice,hemostasis and coagulation . The aim of comprehensive utilization of peanut skin is obtained. The conclusions of the researches were follows:1. The best extraction technology of peanut skin polyphenols'is microwave. The parameters are: The peanut skin grinding particle size is 20 mesh, the alcohol concentration is 63%, the solid-liquid ratio is 1:48, microwave power 750W and time 60s. Test 5 times in these conditions, the extraction amount of peanut skin polyphenol is 152.67mg/g. Compared to the theoretical value, the relative error is about 0.006%.2. From high to low, the content sequence of 8 kinds of peanut skin'total polyphenols is: No.11 luhua, No.1 haihua, No.8 zhonghua, yuanza 9307, yuanza 9102, No.15 yuhua, bairihong. The total polyphenols'content of first four kinds is about 190mg/g and of two yuanza kinds is about 160mg/g.To No.15 yuhua and bairihong, this number is about 130mg/g.3. The conditions of HPLC for the analysis of peanut skin'polyphenols are: Venusil Mp-C18 column(150×4.6 mm, 5um)is used. The mobile phase is as follows: v(CH3CN):v(4mM CH3COONa, Ph=3.0)=15:85, and the flow rate is 1.0mL/min, temperature of column is 35℃, quantity of injection is 10μL.It was measured by an ultraviolet detector under 280.0nm×0.2AUFS.4. With the choose stations of HPLC, the retention time of tannic acid, quercetin, chlorogenic acid, catechin, 4-h-benzoic acid, epicatechin, vanillic acid, vanillin, trans-cinnamic acid, rutin and benzoic acid was determined and the results are: 3.917min,5.258min,6.983min,7.567min,9.808min,10.358min,11.367min,19.258min,25.758min,28.375min and 35.075min。5. The peanut skin'polyphenols of 8 kinds was qualitatived at the same situation and concluded that the peak profiles between the 8 kinds are quite similarly and all kinds of peanut skin polyphenols contain tannic acid, quercetin, chlorogenic acid, catechin, 4-h-benzoic acid, epicatechin, vanillin, trans-cinnamic acid, rutin and benzoic acid and never contain vanillic acid.6. The AA model was proved successful by compared the pathogenic group to the blank group. Between pathogenic group and high,middle dose group, there are significant differences at liver index, spleen index, red cell, white cell, platelet, reticulocyte and bone marrow nucleated cells(BMNC) of mice, this showed that high,middle dose can greatly anti AA.7. Between blank group and high,middle,low dose group, there are significant differences at bleeding time, bleeding amount and clotting time of mice, this showed that all does can decreased the bleeding time of mice, reduced the bleeding amount of mice and shorten the clotting time of mice obviously.All of these displayed the great function of peanut skin polyphenols at hemostasis and coagulation.
