| In this dissertation, the contents of total phenolics, neutral phenolics, acidic phenolics and procyanidin in four apple varieties, including Qinguan, Fuji, Pink Lady and Granny Smith, were determined during development of fruits. The apple polyphenols was purified through macroporous resin and sephadex-LH20. The method was established with HPLC to test the purifid apple pomace polyphenols of Qinguan, Fuji, Granny Smith, Pink Lady, to supply the foundation datas for reserching apple polyphenols in system..The results were as following:1. The contents of total phenolics, neutral phenolics, acidic phenolics and procyanidin in Granny Smith were always more than that in the other three varieties during the whole development(June to October). The other three varietyies, the contents of total phenolics from high to low was Fuji, Qinguan, Pink Lady. The contents of neutral phenolics from high to low was alike with the total phenolics in June, July, Seputember, October. The contents of acidic phenolics from high to low was Fuji, Pink Lady , Qinguan in June, August, Seputember, October. In the ped-fruit period(June), the contents of total phenolics, neutral phenolics, acidic phenolics in Pink Lady was the lowest. They were 62%,57% and 57% of that in Granny Smith. The contents of procyanidin in Qinguan were lowest, only 25% of that in Granny Smith.2. The contents of total phenolics in four apple varieties were decreased during June and July, increased from August and September were highest, decreased during from October. In the early days (June to August) , the contents of neutral phenolics in Qinguan were first decreasing, then increasing, the other varieties were increasing, then decreasing. The contents of acidic phenolics in Granny Smith and Pink Lady were increasing druing development, then decreasing during maturity (September and October), were always decreasing in Qinguanan and Fuji. With the decreasing of the neutral phenolics and acidic phenolics, the contents of procyanidin increasing in the growthing period. The contents of procyanidin was deccreasing in the maturation period (September and October).3. The changes of composition of polyphenolys during development of four varieties was analyzed. The results showed that the proportion of neutral phenolics, acidic phenolics and procyanidin in total phenolics were changed between fruit development. In the early days (July), the contents of neutral phenolics were the main Polyphenols, 74.7% of total phenolics, the acidic phenolics and procyanidin was 13.79% and 11.51% of total phenolics. With fruit development, the neutral phenolics and acidic phenolics were decreasing, but the procyanidin were increasing. The congtents of procyanidin were raised to 46.56% at maturation period, neutral phenolics and acidic phenolics were descended to 41.23% and 12.2%. The other varieties showed identical tendency.4. By captive tests, we detected the absorbing and eluting capacities of three kinds of macroporous absorbent resins producing by two companies. The results indicated that D4020 resin was the best one with higher absorbing and eluting capacities than the other two kinds of resins.5. The purity of Qinguan, Fuji, Pink Lady and Granny Smith after purified through macroporous resin was 23.5%,54.6%,49.8% and 57.6%, the yield was 63.8%,72.5%,62.1% and 79.1%. The purity of Qinguan, Fuji, Pink Lady and Granny Smith after purified through sephadex-LH20 purified again was 85.1%,99.2%,94.4% and 89.3%, the yield was 89.5%,88.4%,78.3% and 73.8%.6. The neutral phenolics HPLC conditions were as follows: column insertsil ODS-3 250*4.6mm, column temperature 25℃, velocity of flow 1.0 mL/min, DAD detector with 280nm wavelength, sample size tenμL, mobile phase acetic acid/ acetonitrile, condition of gradient elution. The acidic phenolics HPLC conditions were as follows: column insertsil ODS-3 250*4.6mm, column temperature 25℃, velocity of flow 0.8 mL/min, DAD detector with 280nm wavelength, sample size oneμL, mobile phase acetic acid/ acetonitrile, condition of gradient elution.7. The results of HPLC detection show that the neutral phenolics all were more than the acidic phenolics. The various kinds of neutral phenolics in Granny Smith were more than that in the other varieties. The contents of the neutral phenolics in Qinguan, Fuji, Pink Lady and Granny Smith were 93.4%, 100%, 98.4% and 99.2% in their total phenolics. The maximum phenolics was Quecetin-3-D-galactoside in four varieties. In the detected total phenolics, the proportion of Quecetin-3-D-galactoside of Qinguan, Fuji, Pink Lady and Granny Smith were 56.06%, 83.91%, 62.69% and 32.26%. The contents of total phenolics detected by HPLC in Qinguan, Fuji, Pink Lady and Granny Smith were 89.5%, 95.6%, 78.86% and 93.28% of their total phenolics before HPLC detection. The other polyphenolics undetected were could other procyanidins and its polymer. |
