Agaricus Blazei Murrill (ABM) belongs to Basidiomycotina, Hymenomycetes, Agaricales, Agaricaceae, and Agariceace. ABM is a kind of precious fungus used for food and medicine, which has rich nutrition and special health value. Extraction technology, the anti-oxidantive effect and the antibacterial activity of ABM polyphenol were studied in this paper. And then the active components of ABM polyphenol were analyzed by Waters 2695 HPLC. Therefore, an important recommedation for developing deep-processing products of ABM polyphenol were offered.(1) An extraction process of polyphenol from dried ABM was studied in this paper. Water and ethanol was used as the solvents and the material studied consisted of dried ABM. Different factors were considered, such as the concentration of solvent, extract temperature, material-water ratio, extract time, and frequency. And orthogonal tests were also done based on the single factor tests. It was indicated that the optional conditions for organic reagents water extraction was: the solidliquid ratio was 1:30, extracting temperature was 90℃, extracting time 3 hours and the frequency was 3 times, the extraction rate could reach to 1.584%. And the optional conditions for organic reagents ethanol extraction were: 80% ethanol solvent was used to extract polyphenol from the dried ABM by 1:25 under 60℃three times and 2 hours for each time, the extraction rate could reach to 2.138%. Compared to the water extraction rate, the Ethanol extraction rate above the method of 35%. It was indicated that the ABM polyphenol contented in the position of hut was 36% more than it contented in the stipe. In addition, ultrasonic and microwave on the extraction rate of ABM polyphenol were studied, the result showed ultrasonic could improve the extraction rate of polyphenol.(2) The extraction and isolation conditions of ABM polyphenol by supercritical CO2 process were studied in this experiment. Influences of indexs which were about extraction temperature, extraction pressure, extraction time were investigated, and suitable conditions were determined based on the single-factor and orthogonal experimental tests. The best extraction technology were: 60% ethanol, extraction temperature 75℃, extraction pressure 40 MPa, CO2 Current capacity 20L/h, the amount of entrainer 120mL. And then compared with static state extraction and dynamic extraction, static state extraction 1 hour, and then dynamic extraction 1 hour was best, the extraction rate could reach to 2.431%.(3) The effect on scavenging·OH and·O2- of ABM polyphenols were studied by Fenton reaction and Pyrogallol Autoxidation Method respectively, which were compared with the effect of tea polyphenols. The result showed that ABM polyphenols could effectively scavenge·OH and·O2- in the experimental range, and the ABM polyphenols had stronger scavenging ability on·O2- and lower ability on ?OH compared with tea polyphenols. In addition, the anti-oxidation of ABM polyphenols was studied. The result showed that ABM polyphenols were effective in lengthening the oil to guarantee the quality.(4) The antibacterial activity of ABM polyphenols was studied, which was compared with the effect of tea polyphenols. It showed that ABM polyphenols had a significantly positive inhibitory activity against Staphylococcus aureus, Escherichia coli, and Bacillus subtilis. But ABM polyphenols had no antibacterial activity against Trichodenna viride and Stachybotrys Chartarum. Meanwhile, temperature changing could not affect the antibacterial activity of ABM polyphenols within limits, and antibacterial effect was not obvious different after changing pH value. In addition, the MIC for three kinds of bacteria were respectively as follow: Staphylococcus aureus, 6mg/mL; Escherichia coli, 3mg/mL and Bacillus subtilis, 4mg/mL.(5) The active components of ABM polyphenols were analyzed by Waters 2695 HPLC. The conditions of chromatography were: reversed-phase C-18 chromatography column, column temperature was 30℃, run-time was 130min, flow rate was 1mL/min, detection wavelength was 260nm, mobile phase was adapted variable gradient with methanol-water solution. And the ABM polyphenols could be analyzed and the Chlorogenic acid could be separated on these conditions.
