Object:To study extraction artwork of epimedium polysaccharides and the immunoregulation role of Epimedium Polysaccharides on the Immune Function of Immune-Depressed Carp,and to provide theory for application of EPS on fish disease field. Methods:1.To compare extracte condition from several aspects such as imbitition,ethanol concentration,extraction frequency.2.To induce carp immune depressed by inject cyclophosphamide by intraperitoneal injection.3.To evaluate white blood cells number,red blood cells number,bactericidal activity of PBL,phagocytize activity of phagocytic cell,activity of serum antalzyme,content of C3 complement and survive rate of Carp that fed with basal diets supplemented different dose EPS after vaccination on time interval after feed experimental group inequality dosage of epimedium polysaccharides in order to analyze immunoregulation function of EPS on immune-depressed carp from both time-effect relationship and dose-effect relationship.Result:1.Had found best extraction condition for of epimedium polysaccharides.the proportion of epimediurn polysaccharides and liquid is 1:20 and repeat 3 times,concentration of ethanol concentration is 70%,and extraction percentage is 5.44%.Epimedium polysaccharides is gained from rude epimedium polysaccharides by Sevag and elution by DAB-6,acetone and ethanol.The content of epimedium polysaccharides is 75.02%.2.Cyclophosphamide had been injected into carp on 5d,10d,15d which dosage is 10mg.kg-1.Evaluating white blood cells number,red blood cells number,bactericidal activity of PBL,phagocytize activity of phagocytic cell,activity of serum antalzyme,content of C3 complement,all data is different from negative control group.Survive rate of the 3rd and 4th experience group is higher than negative group significantly.3.In this study,immune-depressed carp had recovered by be feed bait vessel consist of EPS.3rd group and 4th group had recovered to normal level after be feed 14d.5th group,the HD group(2000mg),WBC,PBL and phagocytize activity of phagocytic cell had recover to normal level but no significant.While number of blood red blood cells,activity of serum antalzyme,content of C3 complement didn't recover to the normal level.The survive rate of 3rd group and 4th group is higher than negative control group significantly.Conclusion:1.Induce immunosuppression succeefully by intraperitoneal inject cyclophosphamide 10mg.kg-1.2.On the study,EPS has regulate significant effect to immune-depressed carp.Immunoregulatory activity related with dosage,which is highest when addict 500mg/Kg,1000mg/Kg EPS into bait vessel.Almost all group had appeared maximum of detection data on 14~21d after feed bait vessel addicting EPS and difffernt with negative control group csignificantly(P<0.05).Continuing fed bait vessel,all of detection data had not increased significantly,indicating that immunoregulatory activity of EPS related with time effect.
