| Zantedeschia aethiopica, a perennial root plant, belongs to the family Araceae and the genus Zantedeschia. It has a close realtionship with Zantedeschia hybrid. Both of them are considerable foliage plants and cut flowers. Relative to the large-scale planting of Zantedeschia aethiopica in China, The seed bulbs of Zantedeschia hybrid are expensive because of long-term dependence on imports. Large amount of seed bulbs for planting, storage and transpot increase the cost and therefor limit its popularization. Tissue cultrue in rapid propagation are uesd to solve these problems in an effective way by now.The rapid propagation of Zantedeschia aethiopica and Zantedeschia elliottiana were systematically studyed to find the effective way, including explants selection and sterilization methods, caespitose shoots induction, proliferation and root induction. By using the strong potential of bud and root induction of stem tuber, studying the relationship between main factors and optimization of technical target, we have established a fairly complete rapid propagation system and analyzed the diversity of two tissue culture processes between Zantedeschia aethiopica and Zantedeschia elliottiana. The main results of our research as follows:1. The stem tuber and leaf of Zantedeschia aethiopica and Zantedeschia elliottiana were uesd as explants. In the condition of common treatment, the best sterilizing time of stem tubers was 13 minutes both of Zantedeschia aethiopica and Zantedeschia elliottiana. But the contamination rate of Zantedeschia elliottiana was still high and unacceptable. By immersing the stem tuber in 50% Carbendazim by 500 times water in 4 hours, or adding 20mg/L penicillin in MS medium had good effect on decreasing the contamination rate. The best sterilizing time of leaves was 6 minutes both of Zantedeschia aethiopica and Zantedeschia elliottiana.2. MS + NAA 0.5 mg/L + 6-BA 2 mg/L + 2,4-D 2 mg/L had the optimal effects on callus inducing form leaf explants both of Zantedeschia aethiopica and Zantedeschia elliottiana. After several transfers of culture, the callus didn't grow vigorously, and the browning effect increased over time. For the purpose of rapid propagation, leaf explants is not an ideal material.3. In the caespitose shoots induction and proliferation experiment, choosing the hormone of NAA and 6-BA to achieve the good induction rate and proliferation coefficient through the quadratic regressive orthogonal design. The results indicated that 0.13mg/L NAA and 1.72mg/L 6-BA in Zantedeschia aethiopica, 0.13mg/L NAA and 1.71mg/L 6-BA in Zantedeschia elliottiana can attain the best induction rate. 0.18mg/L NAA and 1.24mg/L 6-BA in Zantedeschia aethiopica, 0.19mg/L NAA and 1.06mg/L 6-BA in Zantedeschia elliottiana can attain the best proliferation coefficient.4. Adding single hormone in medium can achieve good effects in root induction. In the medium of 1/2 MS + IBA 0.3mg/L, both of the plants could grow more and thicker roots, and became vigorous. |
