In order to investigate the changes of IGF-1,PDGF and BGP throughout the healing of the intramedullary nail fixation of the fractured femur,the relationship between the three cytokines and the recovery of the bone fraction,the changes in X-ray film images and movement function recovery of canine,and the impact of the intramedullary nail fixation on the healing of bone fracture,models of canine intramedullary nail fixation of femoral fracture were constructed.In the study,20 hybridizated canines whose age ranged from 6 to 7 months and weight ranged from 5 to 6 kilograms were selected.Then all animals were grouped randomly by the time points. Each canine had one of the hind limbs chosen randomly to construct the model of canine intramedullary nail fixation of femoral fracture.There were six assigned time point,and at each time point the 20 canines were performed to blood collection and x-ray examination.In this way, the fractured femur's x-ray radiograph,the movement function and the concentration of the IGF-1, PDGF and BGP before and after operation were observed dynamically,and the changes were evaluated.The results showed that:(1)The primary callus formed well on 30th day after operation.The line of the bone fracture basically disappeared on 70th day,but the calcification of the new bone had not been completed at this time.And the callus's moulding were completed mainly on 100th day. (2)The changes of IGF-1 in the serum showed that the concentration of IGF-1 reached the peak at 15 days after operation,and then gradually decreased to the normal level.(3)The changes of PDGF in the serum showed that the concentration of PDGF also reached the peak at 15 days after operation and then gradually decreased to the normal level.(4)The change of BGP in the serum showed that the concentration of BGP reached the peak at 30 days after operation,and then gradually decreased to the normal level.The conclusions are following:During 70th to 100(th) day after operation,the line of the bone fracture of the canines aged 5 to 6 months disappeared,the callus's moulding were completed basically and the fracture ends healed firmly.The intramedullary nails were removaled on 70(th) day. And after that,the movement function recovered well and no bone fracture happened again.In addition,the concentration of IGP-1 and PDGF presented visible increasing at 15 days after operation.This indicates that osteocytes are generating massively which lays the foundation for the primary callus formation and the healing of fractures.The concentration of BGP presented a obvious increasing at 30 days after operation that promotes the calcification of fracture ends and regulates the procession of bone's transition.As a result,the healing of the intramedullary nail fixation operation of canine's fractured femur is identical with the normal healing of canine's fractured femur,it means that there is a little contribution of the treatment of the intramedullary nail fixation operation to the healing of canine's fractured femur. |
