Bornel,Ethyl acetate,Amino acid,Vitamin,flavones are the mainly components of the Chrysanthemun morifolium Ramat..Flavonoids compounds are chiefly a kind of ones whose basal karyomere is 2-phenyl-chromone,and generally these compounds that are plant secondary metabolite that have the basic structure of C6-C3-C6.The flavones compounds that widely exist in the natural plants are principal free or in the form of glycoside.The quantities and species of the compounds are so complicated and so much, simultaneity they are so much pharmacologic activation which can cure the disease of the system of heart,brain,blood vessel and the system of breath,and the same time the Flavones have the pharmacologic activation of resisting the inflammation and bacteria, decreasing the blood sugar,standing up the oxidization,the radiation,the carcinoma and the tumour,furthermore they also can strengthen the ability of immunity.In order to develop and use the Chrysanthemum morifolium Ramat.better,scientists are seeking the valid pharmaceutical which can decrease the blood sugar and strengthen the ability of immunity,so it is necessary that the extraction,purification,identity of the flavones and the deeply study of the biochemistry and pharmacologic activation.They are two parts in the research.The mainly purpose of the first one is foster the callus of the stem tine stripping and the tissue culture techniques.The second one is mainly extract the flavones from the callus of stem tine by two methods,the one method is ethanol extraction,and another one is enzymatic ethanol extraction,comparing the two methods, and at the same time,we take a quantitative determination and qualitative assay of the flavones compounds.In order to cultivate the stem tine callus of Chrysanthemum morifolium Ramat.,The well growing stem tines were chosen.By the sterilization and disinfection,after cleaning by the distilled water,they are inoculated into the good culture media.The effect is that almost all of the stem tines growing well but only some inoculation infecting bacteria,and the well growth stem tines swelled into callus.The induction ratio of the callus is 44.38%, and the callus growth curve of the callus is a type of"S".While the flavones were extracted from Chrysanthemum morifolium Ramat.,the optimum conditions for the extraction were defined by the method of one factor design and the gorthogonal design.That is,firstly,with the condition of the complex enzyme(the enzyme concentration is that cellulose 0.20mg/mL and pentane 0.10mg/mL),enzymatic hydrolysis temperature is 50℃,pH of enzyme solution is 4.5,and the time of enzymatic hydrolysis is 150 min.Then the enzymatic liquid was leached under the conditions which ethanol concentration is 70%,the leaching temperature is 80℃,the leaching time is 120min.Only done this,the effect of the leaching is the best.Compared with the traditional ethanol extraction,both the extraction yield of total flavones and the extraction content of total flavones had been increased greatly.By using the optimum method designed of the enzymatic ethanol extraction,the flavones of the callus from stem tine were leached.The result was as the following, comparing with the stem tine,the extraction yield of total flavones has increased by 34.4%, the extraction content of total flavones has increased by 53.7%.The TLC picture of extraction showed that the chemical components of the extractive have not been changed.To sum up,the new mainly points of this research are as follows:1.The active ingredient contents from the callus of the stem tine have greatly increased.So it could provide a basis experimental base for the incubation of Chrysanthemum morifolium Ramat.2.This study also provided a scientific study method on extracting,isolating, purificating,identifying of flavones from Chrysanthemum morifolium Ramat.Enzymolysis could greatly increase the leaching ratio of the active components in the plants.And this also can provide a basis scientific experimental base for the extraction of flavones efficiently.
