The bulb's scales of Oriental Hybrids 'Tiber' were used as explants and were cultured on MS medium.The effects of IAA,NAA,2,4-D,6-BA and their combination with different concentration on cluster buds,callus and rooting of tissue cultured plants were studied.The aim is to ascertain the best tissue culture conditions of lilium spp,and to provide some technology support for Oriental Hybrids' production.This experiment results indicated that:the medium with 0.5mg/L 6-BA and NAA had a remarkable effects of inducing small scales.There was a cubic functional relationship between the NAA concentration and inducing small scales,Within the experimental concentration of 0.0-0.9 mg/L.The infexion of curve were respectively at(0.36,75.1) and (0.69,131.97).From the experiment results we could known that:it is easy to induce small scale without the addition of NAA,and the inducing rate is 13%.It was explained that maybe there was some growing modulatory substance in the scales.The results of the curve shape also showed that:the number of inducing scales increased quickly with the NAA concentration from 0.15 to 0.36 mg/L.While the number of inducing scales increased slowly with the NAA concentration from 0.36 to 0.69 mg/L,and the highest number of inducing at 0.69 mg/L NAA.The number of inducing scales began to decrease gradually with the increased NAA concentration over 0.69 mg/L.Compared with the control,the number of small scales increased by 11.7 times at the best NAA concentration(0.69 mg/L).The treatment of 2,4-D concentration on callus of the lilium spp.bubils had tremendous effects.The explants could not formation callus below 1 mg/L 2,4-D concentration.While 2,4-D concentration 1.2-3.6 mg/L,between 2,4-D concentration and inducing rate of callus was a quadratic functional relationship.The inducing rate of callus reach to the maximal 60% at 3.2mg/L 2,4-D concentration.With the 2,4-D concentration increasing continually,the inducing rate began to bring down a little.Of these factors on small scales propagation culture,sugar was the largest impact on small scales weight,followed by 6-BA,and then NAA.Low concentration of sugar,6-BA and NAA were good for small scales propagation.The best culture medium for small scales propagation was sugar 30 g/L+6-BA 1.0 mg/L+NAA 0.5 mg/L.1/2 MS medium was better than 1/3 MS medium for rooting propagation.Concentration level of IAA,at the more than 2.0 mg/L or lower than 0.5 mg/L,infected the rooting propagation.The best concentration was 1.0 mg/L,with this concentration,the rooting propagation reached to 93.5%.The 1/2 MS+IAA 1.0 mg/L+IBA 0.2 mg/L+sugar 30 g/L was an appropriate medium for rooting propagation.The detoxification of lilium spp was also performed in this paper.There were great effects of different types and concentration combination of hormones on the inducing ratio of callus,clusters of buds and roots.The best medium for callus inducing was MS+0.5 mg / L 6-BA+0.2 mg / LNAA and the inducing rate was 70.5%;Different medium with the highest rate is MS+BA 1.5 mg / L+NAA0.1 mg / L and the rate comes up to 93.33 percent;the best medium for rooting is 1/2 MS+IAA 0.5mg / L.Stem tips used as a explants for the production of virus-free seedlings can not be greater than 0.6 mm.Stem tips cultured connected with heat dispose is better than only stem tips cultured for detoxification.The appropriate stem tips size is 0.3-0.6 mm.The detoxification rate could reached to 55%.We also preliminarily investigated the aerial root inducing.The method of MS medium culture has the most effect on weight increase and new roots.The average number of new aerial roots is 6.8.So it is possible to improve the survival rate of transplanting.
