Preparation Of Herba Taraxaci-Chamaejasmine Injection

To test their anti-bacteria on S.aureas, E.coli, S.dysagalactie,anti-inflammatory, antipyretic,and to test those pharmacodynamic actions of total flavonoids of chamaejasme;To choose solvent and supplemental agents, and prepaire injection in accordance with the best proportional of the extracts from herba taraxaci and total flavones in chamaejasmine;And to test the impurities, stability, security and the action for the cytophagocytesis of MΦand immune organs of the injection. Experimental methods and results as below:1. The inhibitory action of extracts on S.aureas, E.coli, S.dysagalactie were tested.The results show that the minimal inhibitory concentration (MIC) of the fraction of herba taraxaci eluted with 40% of ethanol (40%HT) on those bacteries is 0.31,0.31,1.25 mg/mL.And the minimum bactericidal concentration (MBC) is 10,10,40 mg/mL;The MIC of the fraction of herba taraxaci eluted with 90% of ethanol (90%HT) is 0.62,0.31,1.25 mg/mL.And the MBC is 10,40,40 mg/mL;The MIC of total flavones in chamaejasmine(TFC) is 0.078, 0.078, 0.16 mg/mL.And the MBC is 2.5,5.0,5.0 mg/mL.And there is auxoaction between 40%HT and TFC. The anti-inflammatory and antipyrecit action of those exacts were designed by swelling of mice pinna by xylen ear and the model of milk cause rabbit heat.The results show that the anti-inflammatory action of TFC was strong and the fraction of herba taraxaci eluted with ethanol had good antipyretic.2. To identify 40%HT:TFC=4:1 as the prescription which were dissolved with ethanol: water: propanediol = 1:4.5:4.5 and added sodium thiosulfate and glycine as supplemental agents to prepair injection.3. For further quality control, the ultraviolet spectrophotometry method of the Neochamaejasmine B was studied. The result showed that in concentration 1.0~10.0 mg/L, the standard of Neochamaejasmine B shows good linearity(R2=0.994,n=6) . The protein, tannin, oxalic acid and resin are checked. The results were in line with the Pharmacopoeia of the People's Republic of China 2005.4. The result of light stability experiments and heat stability experiments showed that the injection can be preserved two years in the condition of 25℃.The injection of the origial, hemolytic, irritating comply with the requirements through experiment. LD50 test results LD50 was 1924.23 mg/kg, showed that the injection was low toxicity.5. Results of the non-particularity immunity tests showed that the injection could increase the ability of macrophage engulfing chicken red blood cells (P<0.01) and could enhance the multiplication of lymphocyte (P<0.01) and thymus at the dose of 100 mg/mL.These experiments had test the main pharmacodynamic actions of HT and TFC, and prepaired injection, which provided datas for extracts and the treatment of mastitis.
