Purpose: The study was designed to research on the establishment of canine hormones avascular necrosis model in order to explore the pathogenesis mechnism and its clinical treatment. The feasibility of the treatment of steroid-avascular necrosis by vascular intervention injection with Fufang Danshen injection and the effect of the treatment would be studied in this paper.Methods: Application of hydrocortisone acetate prednisone in accordance with the 7.5 mg / kg of each dog for intramuscular injection, and each dog will be injected one time every two days by continuous use of four weeks. The weight, serum corresponding indicators and Artery angiography of the 20 dogs of model group would be detected every week, and the dogs would be put to death for the femoral head pathological examination four weeks later and the changes of the model before and after would be compared. The clinical results of the treatment of hormone-avascular necrosis by intervention of Fufang Danshen injection would be studied primarily in the second part of the study. It was divided into 4 groups in this part of study as group A (healthy control group), B (model group), C (treatment model groupâ… ) and D (treatment model groupâ…¡and there were 8 dogs in each group. Group A: intramuscular injecting of normal sodium (NS) with 4 ml each and once a day. Group B: intramuscular injecting of NS with 4 ml each and once a day. Group C: intramuscular injecting of Fufang Danshen injection with 4 ml each and once a day. Group D: rotary Unit within the artery intervention injecting by a rotary unit within the artery intervention of Fufang Danshen injection with 4 ml each and once a week. A series of injections would be last for four weeks of each group.The weight, serum corresponding indicators and Artery angiography of the dogs of each group would be detected every week, and the dogs would be put to death for the pathological examination four weeks later and the changes of the indicators before and after the treatment would be compared each other and the treatment effectiveness analysis would be pointed out.Results:Four weeks after intramuscular injecting of hydride prednisone acetate with 7.5 mg / kg of 16 canine of the model group, it showed that there were no significant changes between the femoral head and the acetabular Waterloo by X-rays of 16 canine while the bone density cut down slightly and it showed that there were not obvious crescent syndromes in part of the dogs. The plasma total cholesterol, triglyceride and low-density lipoprotein have increased, but the high-density lipid protein decreased while the experimental group compared with the control group (P <0.05) and there were obvious differences between the two groups (P <0.05). There were many pathological changes of the model group as the bone trabecula displayed thinning, increased spacing, structural disorder, partly fracture, the majority of cell degeneration and necrosis, marrow fat cells increased and health blood cells to reduce. The vascular diameters were small of the main suppliers for femoral head in model group and the branch of microvasculars were small and thin. The steroid-avascular necrosis model was confirmed of the dogs in experimental group based on the above results. After treatment of the dogs of the second part of the study by intramuscular injection and vascular intervention, the plasma total cholesterol, triglyceride and low-density lipoprotein have decreased of group C by serological examination, and it was significantly that compared between the results of the group C and D and the model group (P <0.05). It displayed that the bone trabecula arranged neatly and the fat cells decreased by bone biopsy. The main vascular suppliers for femoral head were obviously thickening in group C and D, and the branches of microvasculars were also obviously thickening. The treatment results of group D were better than that of group C based on the individual indicators.Conclusion: 16 dogs of the 20 experimental dogs were confirmed to be developed the steroid-ANFH model four weeks after intramuscular injection of hydrocortisone acetate prednisone with 7.5 mg /kg once every two days, and the achievement ratio was 80% and angiography could be used as an early diagnosis of steroid-ANFH. Fufang Danshen injection can significantly lower total cholesterol and triglyceride, low-density lipoprotein increase, and promote the restoration of bone trabecular, and it can also make the major blood vessels supplied to the femoral head diameter thickening, microvascular branches and the blood flow supplied to the femoral head increasing. The treament results of Fufang Danshen injection by rotary Unit within the artery intervention injecting were more obvious than others.
