Clinical Mastitis is one of the common diseases occurs on Dairy Cattle with its strongly influent to Dairy Cattle Breeding Industry.Currently,Cytokine Therapy,which aimed to adjust the function of different immune cells in mammary,enhance its immune performance and improve its immunity to pathogen,due to its advantages,such as easy-produced by DNA recombinant technique,short half-life,less residual in dairy,small dozes and high bioactivities,is becoming the hotspot of recent researches.In the experiment,10~4 cfu/ml/3~4Kg(body weight) of Staphylococcus aureus that was isolated from the milk of dairy cattle with mastitis were infused into the third of mammary gland in 48 healthy rabbits,then the clinical mastitis model were challenged.16 clinical were randomly selected from the 48 groups,observe its manifestations and pathological changes in breast tissue morphology,and regress the breast tissue fluid after established model 72 hours.Another 32 rabbits were randomly divided into A(IFN-γ),B (Ciprofloxacin),C(IFN-γand Ciprofloxacin Lactate),D(normal saline) as four treatment groups,each contains 8 rabbits,then delivery drug only after 6h,18h as first dosage.Each group,respectively,from before established model 1h and 4h,12h,24h,36h,48h,72h observe their temperature,heart rate and respiratory rate and other clinical indicators,and collect blood 0.5ml,Serum 1ml,for blood and blood biochemical detection,and determination of inflammatory cytokines(ELISA),to study its therapeutic effect.The results showed that:1,the model performance(1) Post-challenged treated group expressed significant clinical signs,manifested as reduced dietary wishes,and even waste must;body temperature,heart rate and respiratory rate significantly increased;model swollen breast areas.(2) Challenged district of breast tissue examination found inflammatory cell infiltration,destruction of the organizational structure and so on.(3) Regression tests were positive by both morphological and biochemical identification was identified as Staphylococcus aureus.2,Treatment(1) observation of clinical manifestations,C challenged group started recovering in the 12~24h,after 36h it approached to the pre-challenge level;A,B challenged group started recovering in 36~48h after,after 72h it exceeded the pre-challenge level;D group the extent of infection was positively correlated with the time.Note the use ofγ-interferon alone or ciprofloxacin are able to control their inflammation,and better combination of both.(2) 12~48h after challenged,A,B group,the total number of peripheral white blood cells was significantly higher than the C groups(P<0.01),and was higher than the C group (P<0.05) when the 72h after challenged,but inter-group no significant difference(P>0.05); 12~72h after challenged,D group was significantly higher than the A,B,C group(P<0.01). Results show thatγ-interferon and the combination of ciprofloxacin in the body increased neutrophil phagocytic leukocytes and bactericidal power is superior to other groups.(3) 12~48h after challenged,A,B Serum calcium content was significantly lower than the C group(P<0.01),and was lower than the C group(P<0.05) when the 72h after challenged,However,inter-group difference was not significant(P>0.05);12~72h after challenged,D group was significantly lower than the A,B,C group(P<0.01).Show that theγ-interferon and the combination of ciprofloxacin lactate than alone are more effective in slowing decline in the level of calcium ion concentration,reducing capillary permeability and reduce inflammatory exudation.(4) C group of TNF-αand IL-6 concentration was lower than A,B group(P<0.05) when the 12~24h and 72h after challenged,and was significantly lower than A,B group (P<0.01),A,B groups were not significantly different(P>0.05);the other three groups was significantly lower than the D group(P<0.01) when the 12~72h after challenged.Thatγ-interferon and the combination of ciprofloxacin lactate than the separate use of more effective control of the exudative inflammatory cytokines,reduced inflammatory response, controlling inflammation.Conclusions:γ-interferon with ciprofloxacin in the treatment of clinical Staphylococcus aureus mastitis with a certain degree of synergy.The results of this pilot study for future recombinantγ-interferon treatment of dairy cow mastitis based on the provision of a certain.
