Aphid is one of the major pests that affect cruciferous vegetables yield and quality.The identification and screening of germplasm resources which posses excellent resistant to aphid and the application of somatic cell hybrid can overcome the conventional hybrid incompatibility,obtain the hybrid among different species,and transfer insect-resistant gene from the wild species.It can enrich the diversity of insect-resistant genes of the breeding materials,and realize the innovation of insect-resistant germplasm.The objective of this study is to establish aphid resistance about B.spinescens and M.incana by different methods,such as the free choice of aphids,living in a single feeding,feeding a single isolated room and feeding time,and they can serve as good breeding materials of the insect-resistant gene,also the objective of this study is to research initial mechanism of aphid resistant about B.spinescens and M.incana by scanning electron microscopy and mensuration of internal glucosinolates components.it also study hybrid variation situation by Authenticity identification of the tissue subculture of somatic hybrid of Brassica oleracea and Brassica spinescens,this study want to Determine whether PF197 possess aphid resistance and prove whether somatic hybrid technology may transfer outsanding characters of parents to hybrid progenies.The main results were as follows:1 Aphid resistant identification of wild species1.1 It proved that the wild species Brassica spinescens and Matthiola incana have excellent aphid resistance characteristics,and they can serve as good breeding materials of the insect-resistant gene by different methods,such as the free choice of aphids,living in a single feeding,feeding a single isolated room and feeding time.1.2 It confirmed that the main factor of aphid resistance for the Matthiola incana and Brassica spinescens are leaf structure and indole category glucosinolates 4 ME separately.2 Smotic hybrid tissue culture subculture plants authenticity identification of Brassica oleracea and Brassica spinescens2.1 This study indicated that hybrid characteristics have not been significantly affected after a long-term subculture in vitro and heterogeneous genome has not been completely excluded with relative stability.2.2 Comparing the different identification methods,it was thought that the morphological observation was much simpler and directer,and with enough veracity in most of the cases,but the SRAP molecular marker was more exact and dependable,especially for the exceeding asymmetrical hybrids.3 Resistant to aphid PF197 which had been subcultured in vitro for about one year was identificated,the result showed that PF197 posseses good aphid resistance,compared with control plants.It proved that somatic hybrid technology may transfer outsanding characters of parents to hybrid progenies.
