Radish, a kind of important crucifereous vegetables, is full of nutrition and germplasm resource . The research on the genetic diversity of radish has very good promoting to the appraisal, innovation and the effective utilization of the radish germplasm resource. Glucosinolates(GSs) have important biological activity. To carry out the research work of glucosinolates has important role in guiding the breeding work of radish. It also provid new ideas and evidence for explaining the medicinal mechanism of radish. Glucosinolates markers evaluat genetic diversity of radish germplasm resources from the level of the chemical. It is a new method to study the radish genetic diversity and relationship, which will expand the application of plant chemistry in the study of plant germplasm resources. In this article, the genetic diversity and relationship of 127 radish was analyzed using morphological and glucosinolates markers, combining with the hierarchical cluster analysis and principal component analysis . The main results were as follows:1. Eight individual GSs were isolated and identified by HPLC-MS from the radish germplasm, and the GSs composition and content in leaves and edible roots of radish were analyzed. The results showed that the composition in leaves and edible roots of three radishes were identical, but there were significant differences in content.2. It appraised and analyzed the ten morphological characters of radish, the average multiple index was 0.70 based on the data. The 127 radish idioplasm were divided into 4 kinds using the morphological marker. The results of principal component analysis indicated that the first consideration is characters of root, secondly is color and leaf traits when carried on the radish germplasm classification using the morphological characters.3. 127 radish germplasm were clustered into four kinds based on the glucosinolate marker by the method of hierarchical cluster analysis, and four kinds by the principal component analysis. The result of two kind of taxonomic approach maintained consistent basically. In the principal component analysis, the accumulative variance contribution of the first three principal components reached 89.74%.4. Comparison of the data obtained by morphological marker and glucosinolates maker was made by the method of Mantel test. The r correlation value scope was 0.208-0.241 between the morphological marker and glucosinolates marker.5. Comprehensive analysis of the two kinds of method, the clustering results from morphological characters or glucosinolates marker were associated with the morphological characteristics. Finally the 127 radish idioplasms could be classified into three kinds: long, short and flat round radish.
