This paper researched on the method of determining cell wall substance of tobacco.In this foundation of determination method,the contents of cell wall substance of flue-cured tobacco in our contry were analysed,the influces of cell wall substance on tobacco quality were also analysed,the results were as follows:1 With the microwave technology,the benzene,the mellow processing stage hypothesis microwave power was 180W,heated up 30 minutes,the water treatment stage hypothesis microwave power was 240W,heated up 20 minutes,the determination result is consistent with the conventional method.The pectine was hydrolisised into the pectic acid under the alkalinity condition,then produced pectic acid calcium precipitation with CaCl2.To obtain the content of pectine,we could make the content of Ca in pectic acid calcium with ICP multiply 12.072,thus indirectly determined the pectine content,and the determination result was more ideal.To determinate the content of cellulose with acetic acid-nitration,the coefficient of variation of the experimental result is 3.57%.To determinate the content of hemicellulose with 2mol/L hydrochloric acid water solution, the coefficient of variation is 0.74%.So these two determination methods were feasible. 2 The content of total cell wall substance,total cellulose,lignin,pectine,hemicellulose,cellulose were in the ranges of 24%~35%,10%~16%,0.5%~4%,5%~9%,2%~5%,5%~9%respectively of flue-cured tobacco leaf in our country.The components of cell wall substance in cutter leaves were higher in Hunan Guiyang,middle in Guangdong Nanxiong and Fujian Sanming and the content was close with Brazilian and Zimbabwe,lower in Sichuan Liangshan and Panzhihua,Yunnan Yuxi.In the varieties of K326,NC89,yunyan85,yunyan87,zhongyan100,zhongyan 101,the contents of total cell wall substance,total cellulose,lignin in K326 were higher than other five varieties and the content of pectin was lower than other five varieties.The contents of total cell wall substance and pectin were the highest in the lower part leaf,then in upper leaves and lower in cutters.The content of total cellulose was the highest in the upper leaves,next in cutters,lowest in lugs;the content of lignin was highest in lugs,next was cutters, upper leaves were lowest,also remarkably was lower than the lugs and cutters.The content of cell wall substance maturity was close with mature.Total cell wall substance content of mature leaf tobacco was lower and the vacation ripe leaf was higher.The content of total cell wall substance,total cellulose and lignin in lemon of lower and upper leaves was higher than that in orange,in contrast with the cutter;The content of pectine in lemon was higher than that in orange.The content of total cell wall substance and total cellulose with different status was not remarkable and pectin and lignin were remarkable.The pectine content was lowest by the status medium leaf tobacco and the lignin content reduced from thinly to thick.The content of total cell wall substance,pectine,and lignin reduced from loosen to close,but the content of total cellulose increased.The contents of pectin and total cellulose with different content of oil were not remarkable.The contents of total cell wall substance and lignin reduced from low to high and the difference was remarkable.The content of lignin chromaticity enhancement reduced with the increase of color and the contents of total cell wall substance,total cellulose and pectine are not obviously changing. 3 The cell wall substance of tobacco leaf and the index of tobacco quality were analysed,and the partial correlation analysis of the obtained data was carried out,the results showed that:The content of total cell wall substance of tobacco leaves was negatively correlated with tobacco body, tensile load,total volatile base(TVB),total nicotine,quality of aroma significantly,but positively correlated with equilibrium moisture content significantly;The content of total cellulose of flue-cured tobacco was positively correlated with tobacco body,thickness,total volatile base(TVB),total nitrogen and total nicotine significantly,but negatively correlated with reduced sugar and starch significantly;The content of pectine was negatively correlated with the degeneration of aromatic amino acids significantly;The content of hemicellulose of flue-cured tobacco was positively correlated with tobacco body significantly;The content of cellulose of flue-cure tobacco was positively correlated with leaf structure significantly,but negatively with tobacco body significantly.4 The correlation between chemical components,total cell wall substance of flue-cured tobacco and tobacco quality were analysed by grey incidence analysis,the results showed that, Carbohydrates had greater effect on mature and leaf structure,followed by the pectine,cellulose, K+;Nitrogen Compounds had greater effect on tobacco body,oil and leaf color intensity,followed by cellulose and starch.Carbohydrates,pectine,cellulose and SO42-had greater effect on equilibrium moisture; Nitrogen Compounds had greater effect on thickness,leaf density,tensile load and tensile strength, followed by total cell wall substance and cellulose;Lignin,K+,SO42-and hemi-cellulose had greater effect on filling power.Carbohydrates,pectine,cellulose had greater effect on quality of aroma;Carbohydrates and cell wall substance had greater effect on after taste,offensive odor,combustibility and smoking scores;Volatile base,total nicotine and carbohydrates had greater effect on volume of aroma; Lignin,cellulose and pectine had greater effect on irritancy and ash color.
