Gongdejian—an irrigation lavage solution successfully developed by our research group had been used to treat the endometritis of dairy cows, which was based on the modern research materials of Chinese herbal medicine and effectively medical prescription.In order to further improve the prescription and curative effects of the drug, the research modified the old prescription and recombinated two improved prescriptions—prescription I and II. The acute toxicity, hypersensibility, irritative reaction, anti-inflammatory effect, minimal inhibitory concentration in vitro, effect of medication in goat suffered the experimental endometritis treated with improved prescriptions were respectively studied. The paper included 4 experiments as following.1.The security experiment: Feeding mouse with the maximum concentration and the maximum capacity medicine by intragastric administration,the LD50 was not appear, indicating that prescription I and II showed safe in acute toxicity. In the maximum tolerable dose (MTD) experiment, the results were MTD>40g/㎏. No allergic reaction presented in cavy during allergic testing and no hyperemia was found in the conjunctiva during stimulation test.The results testified that prescription I and II show no stimulation and safety for uterus lavage.2.Anti-inflammatory test and bacteriostasis experiment: The result of anti-inflammatory test indicated that the middle dosage group (10g /kg body weight) and the high dosage group (20g /kg body weight) of prescription I,the middle dosage group (10g /kg body weight)of prescription II could obviously inhibit the swelling of mouse'ears which were caused by xylene, while the high dosage group of prescription I could inhibit the swelling of mouse'ears extremely significantly which were caused by xylene, the high dosage group (20g /kg body weight) and the middle dosage group (10g /kg body weight)of prescription I and the high dosage group of prescription II could obviously inhibit the accrementition of granulation tissue caused by cotton balls. Compared prescription I or II with the old prescription, the anti-inflammatory effect of prescription I was the best one. The bacteriostasis test in vitro showed that the minimum inhibitory concentration (MIC) of prescription I to the E.coli,Staphylococcus,Pseudomonas and Streptococcus were 0.0125,0.125,0.1,and 0.25g /ml, respectively, but prescription II were 0.025, 0.1, 0.25, and 0.25g /ml, respectively. Compared prescription I or II with the old prescription, the effect of bacteriostasis of prescription I was the best one.3.The establishment of models of endometritis in goats. The E.coli isolated and purified from clinical case suffered endometritis was applied in the experiment to establish the model of endometritis. The goats were randomly divided into 2 groups (group I and groupII), each one with 6 goats. Through pipe which was made by the uterine horn intubation, the cavity of uterus was infused with sterile normal saline in group I, while the E.coli (O18, 5×108CFU) was inoculated in the cavity of uterus of groupII. E.coli was inoculated with additional 5×108CFU on the original concentration until the goat excreted purulent secretion at a 48h-interval.The results showed that the goats of group II excreted purulent secretion after 6-7th days of the second bacteria inoculation. The secretion increased after 8-9th days. The methods might successfully replicated the disease model of endometritis in goats.4.Therapeutic test of endometritis with improved Gongdejian prescriptions- prescription I and II in goats. 24 goats with endometritis caused by E.coli were randomly divided into 4 groups. In group IV, 20ml of normal Saline was infused into the cavity of uterus by uterine horn intubation. By the same method, the goats uterine cavity were infused with 20ml of physic liquor prescription I in grope I, physic liquor prescription II in grope II, 1% ciprofloxacin solution in grope III, respectively , one time at a 2d-interval.After 3~4 times medication, the general condition and physic-chemical characteristics of discharges were recovered, the therapeutic effects were better in grope I than that in gropeⅡand grope III.
