LanZhou Lily(Lilium davidii unicoior Cotton)is mainly reproduced in stem bulblets and scales,in this process,the virus can be spread easily,which affects the yield and quality of LanZhou Lily.Virus free and rapid propagation technology could not only free virus,but also improve the quality and propagation rate of LanZhou Lily based on tissue culture.In this paper,the main virus kinds injured LanZhou Lily were detected with the methods of field investigation,double antibody sandwich electric microscope,and Double antibody sandwich enzyme—linked immune sorbent assay(DAS-ELISA);The virus-free trial was done by using material of meristem-tip,filament,petal,scale,seed,meristem-tip and filament with heat treatment.Also,the study about transplant environment,practice methods,transplant medium, time of tube plants and manage conditions after transplant were systematic done.1.At present,the primary virus injured LanZhou Lily were CMV,LSV,TBV,and infested mode was compound and recessive.2.The meristem-tip culture with heat treatment could improve the efficiency of virus-free,the elimination rates was 70%;The efficiency of virus-free by Filament culture with heat treatment was the best,the elimination rates was over 80%,this technology was simple,speedy and efficient,and could apply in the production of LanZhou Lily.3.When used filament as explants,the contamination rate was low with sterilizing time for 15-20 minutes.Considered the growth,color,direct differentiation numbers of tube plants,the optimal medium were MS+BA1mg/L+IBA0.5 mg/L for induction of filament, MS+BA1.5mg/L+NAA0.2~0.5mg/L for subculture on which the plant grew green and well,the best medium for root were B5+IAA0.2mg/L with 90.5%root generation rate and vigorous roots.4.The technology for propagation of virus-free initial cormel:in the middle of May,the vitroplant with roots were practiced in a higher light intensity(1.6×1041ux~2.4×1041ux)in a sealed tube about two weeks,then they were cultured in unsealed tube for two or three days.The vitroplants were transplanted to sterilized pasteurized peat placed under insect prevention net with normal light and high humidity and low temperature to propagate initial bulbs of LanZhou Lily. This technology could simplify the transplant process,shorten the circle of initial bulb culture,and low save culture cost.
