Recently,virus diseases are becoming the bottleneck problem of the healthy development of production which cause grave effects on lily.Therefore,the rapid and sensitive technique of virus detection,deprivation and the cultivation of the virus-free bulbs will catch an actual value to lily.In this research paper,5 lily variations were selected to study the detection and elimination technique of virus on Lily bulb.All results of the study were summarized as follows:1.The method of RNA extraction was improved,the results showed that Trizol method was only fit for obtaining RNA from inner scale and CTAB method could used for middle and outer scale.3 sets of specific Primers were designed and the augment conditions were optimized to RT-PCR technology on bulbs and tube plant.2.Five virus-free methods are callus culture(Ⅰ),shoot tips culture(Ⅱ),heat treatment combining with shoot tips culture(Ⅲ),virazole combining with shoot tips culture(Ⅳ) and shoot tips culture combined with ribavirin and heat treatment(Ⅴ).On the comparison of the efficiency and feasibility of the five methods above,the methodⅤis the best,with the methodⅢ,Ⅳcome to the next.3.The advanced and perfect technology for propagation of lily virus-free bulbs was summarized as follows:the optimal intensity of illumination in transition duration for 'Siberia','Sorbonne' and 'Acapulco' is 2.4-3.0×10~4lux and 3.2-4.0×10~4lux for 'Yelloweed',Conca D'or' in a sealed tube about 2 weeks,then they were cultured in unsealed tube for 2-4 days.The vitroplants were transplanted to the substrate matured by peat to vernmiculite to rotted cattle manure 4:4:1.
