This article aims to determine distribution of beta-carotene and Lutein in Sanhuang chicken' tissues and variance in serum by isotopic trace of 14C.In the first experiment, the biological synthetic method of 14C-β-carotene (14C -βc) and 14C-Lutein had been established. The Spirulina maxima was grown in Zarrouk's medium using initially NaHCO3 and subsequently 14C-NaHCO3 enriched medium (specific activity 25mCi/L). After suspension cultured in normal Zarrouk culture medium for producing maximum biomass and carotenoids, Spirulina was kept in radioactive culture medium 2 months in which some C atoms of the Spirulina was replaced by 14C atoms of the culture. Then, βc and Lutein labeled by 14C could be extracted from the Spirulina by chromatogram. The radioactive culture medium could be used in several cycles for metabolic labeling of carotenoids whereby the cost of production was reduced. At last, purity of 14C-βc was about to 95% and purity of 14C-Lutein was about to 80%.In the second experiment, Sanhuang chicken was poured in 14C-βc or 14C-Lutein respectively, by oral to observe responses of serum. Radioactive concentration in serum of chicken poured by 14C-βc peaked at 6h, 20h, 38h and subsequently declined rapidly. By contrast, radioactive concentration in serum of chicken poured by 14C-Lutein has been attained to the peak after 30h and unchanged again after 34h.In the third experiment, the rate of metabolization of 14C-βc was 64.2% and the rate of 14C-Lutein was 62.8%, which was counted out by pouring 14C-βc or 14C-Lutein into Sanhuang chicken. Blood, bone, feather and so on in chicken was accumulated 34.6% of radioactivity from 14C-βc and 29.4% from 14C-Lutein. Small intestines and liver was the major segment for 14C-βc, however, 14C-Lutein was found in muscle.In the last experiment, radioactive concentration in gall was higher (P<0.01) than other tissues significantly, which was non-significantly differed between 14C-βc and 14C-Lutein. Radioactivity in small intestines from 14C-βc was higher than 14C-Lutein visibly, on the contrary, the radioactivity saturation in shank, skin, muscle and fat from 14C-Lutein was higher.
