Sihuang Injection (SHI) was applied to the rabbit's heat syndrome model by injecting the Escherichia coli endotoxin and pigs with fever, the body temperature of the rabbits and pigs was collected according to the schedule, as well the blood of rabbits for the test of IFN-a, PGE2, c-AMP and NO concentration in the blood serum,be aimed at studying the clinical effects and mechanism, The results can be summarized as follows:(1) Establishing the rabbit's heat syndrome model:0.5 hours after the injection of endotoxin, the rabbits of model group began to show various symptoms including hair sticking up, crouching, shivering, decreasing of activity, external ears becoming white and etc. Around 1 to 3 hours later, the body temperature started to increase; the heart beating and respiratory rate increased; the eye congestion showed up; frequency of drinking increased. After that, body temperature began to drop and the above mentioned symptoms disappeared, About 6h returned to normal.This justifies the success of reproducing the rabbit's heat model.(2) The reducing fever experiment for rabbit's heat syndrome model:36 rabbits were randomly divided into 6 groups, with 6 rabbits in each group, including normal control group, model group, Comparison products group, SHI low dose group, SHI medium dose group and SHI high dose group. Except the normal control group, all the other groups were injected with endotoxin through external ear vain to establish the heat syndrome model, which was followed by the injection of the drug. During the experiment, body temperature was collected every half an hour. After three hours observation, the blood was drawn from each group. The ABC-ELISA method was utilized to measure IFN-α,PGE2,c-AMP and NO concentration in the blood serum. The results indicates that the effect of reducing fever in the high dose group is the best, which is significantly different from that in the model group(p<0.01). For the high dose group, the IFN-a and PGE2 concentration in the blood serum decreased significantly (P<0.01),the NO concentration increased (P<0.05) and c-AMP showed no change (P>0.05).(3) The clinical effect for the pigs with fever: the pigs were divided into normal control group, amidopyrinum Injection group and SHI group. Among each group, the pregnant pigs, pigs at confinement stage and pigs at growing stage were treated separately. The results indicates that at each stage, the temperature of amidopyrinum Injection group and SHI group decreased significantly (P<0.01),and the pigs'clinical symptom of fever fade away.The above results indicate that SHI shows significant clinical effect on the rabbit's heat syndrome model by injecting the Escherichia coli endotoxin, and the reducing fever mechanism may be by decreasing the IFN-a and PGE2 concentration and increasing the NO concentration in the blood serum. And SHI can reduce temperature to the pigs with fever in clinic,it has good effect on pigs.
