For the past few years, Livestock and poultry breeding industry appeared a lot of febrile diseases, which give Livestock and poultry breeding industry great influence. Traditional Chinese veterinary medicine thinks, febrile illness all that can be used by wei qi camp blood syndrome differentiation and treatment which founded by qing dynasty physician YeTianshi.Among them, Heat Accumulation of Intestinal Type of Qi-fen Syndrome is one of the important stages in the process of warm disease development, The morbidity animal clinical with fever, intestinal dry and excrement dry, constipation, urine short and red, body fluid in the mouth dry, mouth’s color dark red, tongue coating yellow and thick, pulse heavy and powerful as the main characteristics. At present, There is no mature animal model method on Heat Accumulation of Intestinal Type of Qi-fen Syndrome, And there is no systematic study on animal intestinal mucosal immune function change in Heat Accumulation of Intestinal Type of Qi-fen Syndrome, which bring great difficulties on rebrile disease mechanism and traditional Chinese medicing research ande development.For this reason, the study based on perfuse heat sexual medicine and their dung, intraperitoneal injection inactivate escherichia coli, set up the Heat Accumulation of Intestinal Type of Qi-fen Syndrome animal model; and carry through pathology evaluate On Heat Accumulation of Intestinal Type of Qi-fen Syndrome’s structure and function changes.Determine the intestinal tissue pathological structure change, the related cytokines mRNA expression situation, in order to determine the changes of the Heat Accumulation of Intestinal Type of Qi-fen Syndrome’s function.1. Heat Accumulation of Intestinal Type of Qi-fen Syndrome Can be build by the following conditions: Give rat monkshood, cinnamon, rhizoma zingiberis(1:1:1)12d, after continuous drench their own feces, then intraperitoneal injection inactivate escherichia coli, after5h, rats can apear Heat Accumulation of Intestinal Type of Qi-fen Syndrome symptoms:The whole body shaking, shrug fur, crispatura, Spirit ennui, conjunctival injection, polypnea, heart rate speed up, drinking water times increase, uring reduce and yellowing, defecation time prolonging, the defecation’s grain number decreased, stool stem node, and ball round bead shape or beads, auricle vascular congestion and expanding, increased body temperature and weight loss etc.2. Heat Accumulation of Intestinal Type of Qi-fen Syndrome rat model’s intestinal pathological evaluation is as follows:Intestinal tissue paraffin section and H E staining results showed that:At5h, rat duodenum, jejunum and ileum were all appear significant pathological damage, such as the duodenum had significant inflammatory cell infiltration, and lamina propria and cortex occurred vacuoles, jejunum and ileum mucosa fall off, inflammatory cells infiltration etc other pathological damage;at the same time, duodenum, jejunum, ileum were all appear significant pathological changes, among those, jejunum tissue damage in the most obvious, and further determines that Heat Accumulation of Intestinal Type of Qi-fen Syndrome rat’s intestinal structure severely damaged, especially jejunum.The intestinal mucosa IEL number show:at the5h and7h, IEL number in duodenal were significantly lower than control group (P<0.05) and at7h, duodenal IEL number is significantly higher than5h (P<0.05); At5h, jejunum IEL number was significantly lower than control group (P<0.05), the rest of the group and the control group had no significant difference (P>0.05).Intestinal mucosa GC number shows:the model group rats in the duodenum and jejunum bowel, GC quantity change trends are basically identical. When at5h and7h, duodenal IEL number were significantly lower than control group (P<0.05) and7h, duodenal IEL number is significantly higher than5h (P<0.05); At5h, jejunum IEL number was significantly lower than control group (P<0.05), the rest of the group and the control group had no significant difference (P>0.05).3. Detect Heat Accumulation of Intestinal Type of Qi-fen Syndrome rat intestinal structure cytokines mRNA quantity expression shows:Through the Real-time PCR to detect IL4, IL-10, TGF-p, TNF-a, IFN-y, IgA mRNA expression, found that in addition to the IL-10had highest expression quantity in ileum and colon, the rest cytokines are expressed in the duodenum is the most sensitive.In the condition of Qi-fen Syndrome, Thl type cytokines’s expression change are: IFN-y mRNA expression in addition to the colon, duodenum, jejunum and ileum IFN-y mRNA expression in enhancing; TNF-a mRNA expression quantity at5h group of duodenal expression quantity minimum, jejunum and colon tissue at5h group mRNA expression of TNF-a quantity highest; The ileum with5h group of TNF-a mRNA expression quantity is lower than the control group,3h group; Th2type cytokines expression changes of:IL-4mRNA expression quantity in the intestinal tissue were lower; IL-10mRNA expression of the duodenum and the control group had no significant difference in5h group, the ileum, jejunum and colon expression enhanced.In addition, IgA mRNA which lift in5h group in the duodenum, jejunum and ileum, colon tissue; And in duodenum and colon tissue TGF-β mRNA expression quantity increase in5h group. Jejunum and ileum tissue in5h group, TGF βmRNA expression quantity decreased.To some up, Through perfuse rat heat sexual medicine and their dung, after intraperitoneal injection inactivate escherichia coli five hours, all kinds of clinical symptoms in Rats conforms to Heat Accumulation of Intestinal Type of Qi-fen Syndrome’s syndrome characteristics, namely successfully established Heat Accumulation of Intestinal Type of Qi-fen Syndrome animal model.Under Heat Accumulation of Intestinal Type of Qi-fen Syndrome state, the rat intestinal organizational structure damaged, with jejunum tissues damaged the worst; and IL-4, TGF-β, TNF-a, IFN-y, IgA mRNA expression quantity highest in duodenal.In jejunum and ileum, IL-10expression quantity highest, and the organization of Thl/Th2cells in the immune balance direction to Thl cells, lead to immune imbalance, tissue inflammatory reaction is aggravating, and increased expression such as IgA, embodies the Heat Accumulation of Intestinal Type of Qi-fen Syndrome animal intestinal mucosal immune increased, but has the characteristics of the inflammatory reaction is aggravating trend.
