| In order to offer the theory basis of conversation and utilization of indigenous goat breeds genetic resource. Fluorescence PCR was applied to investigate the genetic diversities of 9 indigenous Chinese goat breeds and 1 exotic breed with 10 microsatellite DNA markers recommended by the Food and Agriculture Organization of the United Nations and the International Livestock Research Institute of Animal Genetics, which provide datas for the conversation and utilization of indigenous goat breeds genetic resource. A total of 447 individuals were detected and amplified products were analyzed based on ABI3130XL auto gene sequenator.(1) Based on the 10 microsatellite loci, we got 119 alleles and 13.7 in average. The most allele number was 19 for TCRVB6, the least was ETH10, which is 6, and other loci were from 11 to 18, and the effective allele number ranged from 1.4641 to 9.2911. In ten goat breeds, the average allele number was 4.6 for Boer, except Fengqing hornless black, other indigenous Chinese goat breeds were higher than exotic breed.(2) At 10 loci the expected heterozygosity was 0.5128 from 0.691; the observed heterozygosity was 0.4224 from 0.6929; and the PIC was 0.4730 from 0.6716. In 10 microsatellite loci of study, one was low polymorphic, one was moderate polymorphic, another eight were high polymorphic, that could be used to the analysis of genetic diversity in goat.(3) The genetic distance between 9 indigenous Chinese goat breeds and 1 exotic breed was larger. The genetic distance among indigenous Chinese goat breeds was smaller relatively (DA=0.0750-0.3972, Ds=0.0297-0.5771), thereinto it was the smallest between Hexi cashmere goat and Xinjiang (DA=0.0750, Ds=0.0297), and it was the bigger between boer and indigenous Chinese goat breeds(DA=0.3458-0.6340, Ds=0.4008-1.1549).(4) We made a synthesis of NJ and UPGMA cluster, the PC A analysis, and the result of Structure program, showed that 10 goat breeds were clustered into four groups. The first group included Xinjiang, Chaidamu, Shannanbai, Hexi cashmere, Fengqing hornless black, the second group included Longling goat, Maguan hornless, Jianchang black, the third group included Banjiao, the forth group included Boer. The result was generally in accordance with their original breeding history and localities. |
