Objects:The relationship between Wnt / β-catenin pathway and TGFβ1-Smad signal transduction during the epithelial-mesenchymal transition of cholangiocarcinoma cells were discussed by observing the changes of factors in TGFβ1-Smad signaling pathway when Wnt2 b induced the epithelial-mesenchymal transition of cholangiocarcinoma cells.Methods: Transfecting human cholangiocarcinoma RBE cells by Wnt2b-si RNA lentiviral vector,β-catenin-si RNA lentiviral vector and empty lentiviral vector to get Wnt2 bsi RNA-RBE cells group which Wnt2 b is suppressed,β-catenin-si RNA-RBE cells group which β-catenin is suppressed and the negative control group.Wnt2 b and β-catenin expression was detected by RT-PCR.At the same time,the expression of Ecadherin,Vimentin,TGFβ1,smad4,snail and slug at the m RNA and protein levels were detected by RT-PCR and Western Blot.Results:Compared with the negative control group,the expression of Wnt2 b in Wnt2 bsi RNA-RBE cells is lower(P﹤0.05),and the expression of β-catenin in β-catenin-si RNA-RBE cells is lower(P<0.05).In the last two groups,the expression of Ecadherin is higher,but the expression of Vimentin is lower(P<0.05).Compared with the other two groups,the expression of TGFβ1,smad4,snail and slug in Wnt2b-si RNARBE cells are lower(P<0.05).On the other hand,compared with the negative control group,the expression of these four factors is the same(P>0.05).Conclusion:Wnt2b-si RNA lentiviral vector and β-catenin-si RNA lentiviral vector can effectively inhibit the expression of Wnt2 b and β-catenin in RBE cells respectively,and inhibit the EMT process of RBE cells by increasing the expression of E-cadherin and decreasing the expression of Vimentin.Wnt2 b can induce the EMT of cholangiocarcinoma cells through the Wnt/β-catenin pathway,but also can exert this effect by TGFβ1-Smad signaling pathway.There was a shaft coupling relationship between Wnt/β-catenin pathway and TGFβ1-Smad signaling pathway.
