Objective: To investigate the temporal and spatial expression of Wnt2b/β-catenin/c-myc signaling pathway in mouse embryonic heart. Methods : The mouse embryonic hearts,which of the sex of fetuses at gravidity 10d(G10d), G12.5d, G15d, G17d, G18.5d, and postnatal day(P0d) , were got ready. Immunohistochemistry, Reverse transcription PCR and Western blot were used to detect the expression of Wnt2b,β-catenin and c-myc during different periods of mouse embryonic heart development. Results: With the progress of the embryonic heart development, the expression of Wnt2b,β-catenin and c-myc at the mRNA and protein levels is decreasing gradually, and the comparison of mRNA expression of Wnt2b,β-catenin and c-myc shows that they are interrelated statistically (P<0.05). Conclusions: These results suggest that Wnt/β-catenin/c-myc signaling pathway plays important roles in regulating the development of the embryonic heart. And it can be predicted that c-myc may be one of Wnt2b downstream target genes. |
