"Wnt" was a combination of the Drosophila segment polarity gene Wingless and the mouse proto-oncogene Int-1.The Wnt extracellular signaling pathway is a very conserved signal transduction during animal development, from Hydra to human. Wnt family is a family of sevreted proteins, which have function in cell growth, cell differentiation, patterning in embrogenesis in organ systems. There are19WNT genes, some of them have alternatively spliced isoforms. On the basis of their acivity in vitro or vivo function model, Wnt proteins have been classfied into two groups, classtic canonical and noncanonical protein.Wnt has got close attention as its strong controlling cell survival, cell fate, and cell movement in developing embryos. After that, Wnt/catenin pathway can control a wide range of cancers development through changing cell differation and cell proliferation, including tumor development, including colorectal cancer, gastric cancer, tumors derived from hepatic tissue, ovarian cancer, breast cancer and prostate cancer. Numerous studies proved that Wnt plays an extremely important role in tumor development and metastasis.WNT2B/WNT13was the thirteenth cloned WNT gene, and then found highly conserved in ouse, chicken, and zebrafish. Human Wnt2B and Wnt2have66.9%similarity, while mouse Wnt2b shows71.1%identity with mouse Wnt2. Frizzled (FZD) receptor is a canonical WNT transduce signalling receptor, and Wnt2B can function as a canonical WNT through this patyway. Meanwhile, Wnt2B can also function as a noncanonical WNT through Ca pathway. Studies have found Wnt2B has expression in fetal lung, brain, kidney, adult brain, placenta, lung, heart, proatat, ovary, colon, as well as some types of human cancer, such as cervical cancer, breast cancer, basal cell carcinoma and leukemia. The highly expression of WNT2B in tumors suggested that there might be some relationship between this gene and tumor development, but the mechanims are limited.Our study used ovarian cancer cell lines A2780,C13K and breast cancer cell lines MCF7,MDA-MB-231as cell model, suggested that WNT2B pathway has important function in metastasis and drug resistance of ovarian cancer as well as development of breast cancer. Its function strongly provided evidences that Wnt2B could be used as a new target for ovarian cancer and breast cancer gene therapy.
