Inhibitory Effect Of Qingre Jiedu Huazhuo Tablet On EV71 Virus And Its Mechanism

Objective:To observe the enterovirus71(EV71) inhibitory effect and effective ways of Qingre Jiedu Huazhuo tablet,and explore the regulation and control effects of Qingre Jiedu Huazhuo tablet in the inflammatory response caused by EV71.Methods:With help of cell culture to observe the anti-EV71effects of Qingre Jiedu Huazhuo tablet, and its incomplete prescriptions of normal cell control group and the virus control group, Ribavirin in control group, Qingre Jiedu Huazhuo tablet control group, In preventive-inhibiting-virus experiment, therapeutic-inhibiting-virus experiment and preventive-therapeutic-inhibiting-virus experiment. To observe changes of expressions of TLR4mRNA and NF-KBp65mRNA in RD cells in inhibiting-virus-directly experiment with intervention of Ganlu Xiaodu Dan, Qing prescription and Li prescription by method of RT-qPCR.Results:1) Preventive experiments, Qingre Jiedu Huazhuo tablet control group cell survival rate significantly greater than the virus control group (P<0.01), the virus growth inhibition rate was48.46%. Therapeutic experiment, Qingre Jiedu Huazhuo tablet control group cell survival rate is greater than the virus control group (P<0.01), the virus inhibition rate was31.05%. Prevention and treatment of sexual experiment, Qingre Jiedu Huazhuo tablet control group cell survival rate significantly greater than the virus control group (P<0.01), the virus inhibition rate was67.67%. three kinds of experimental scheme,Qingre Jiedu Huazhuo tablet control group compared with ribavirin group cell survival rate, and virus inhibition rate, there is no obvious difference, no statistical significance (P>0.05). About Qingre Jiedu Huazhuo tablet control group cell survival rate, and virus inhibition rate, mutual comparison between three kinds of experimental scheme, minimum therapeutic experiment, prevention and treatment experiment, the highest is highly statistical significance between groups (P<0.01).2)The virus control group compared with control group of normal cells, TLR4mRNA, NF-kappa Bp65mRNA level significantly increased (P<0.01). Compared with the virus control group, Qingre Jiedu Huazhuo tablet control group significantly lowered TLR4mRNA, NF-kappa Bp65mRNA expression (P<0.01).Conclusion:1) Qingre Jiedu Huazhuo tablet has good effect of against EV71virus and protective cells, And Through its adsorption, copy by influencing the EV71, improve cellular immune way have the effect of inhibitting the growth of EV71.2) Qingre Jiedu Huazhuo tablet can pass down TLR4mRNA and NF-kappa Bp65mRNA expression, to fight inflammation caused by EV71,that prompt TLR4in EV71infected cells play a role of raising, and activate the NF-kappa Bp65promote inflammation.
