Purpose:In this study by observing the electroacupuncture of "Zusanli","Sanyinjiao"of spleenn deficiency rats colon tissue of vasoactive intestinal peptide (VIP),nitric oxide (NO) and vasoactive intestinal peptide receptor2(VIPR2), nitricoxide synthase (NOS)(SCF) content.Evaluation of acupuncture SCF/c-kit signalpathways regulated influence on gastrointestinal dynamics,and study itsinfluence on inhibitory neurotransmitter and recepterreceptor;Evaluation ofelectroacupuncture of "Zusanli","three vaginal intercourse" of spleendeficiency rats gastrointestinal disorder to improve the effect and its mechanismof action;Discuss acupuncture point adjustment spleen-deficiency type thespecificity of gastrointestinal disorders.Material and method:Using SPF rats, male,7weeks,40, is provided by liaoning university oftraditional Chinese medicine laboratory animal center, randomly divided intonormal group, the acupuncture point group, without going through the acupuncturepoint group, and the model group, both for regular feeding, do not do anyprocessing in addition to the normal group, the other three groups were giventemper deficiency syndrome model building, building for two weeks, exhaustionswimming25minutes per day, the water temperature of36℃to37℃, in threedays for a cycle, two days before fasting, feeding on the third day, drinkwater.Building after the acupuncture point group and cupping treatment withoutgoing through the acupuncture point group, for a week, during the treatment,continue to the model group, the acupuncture point group, without going throughthe acupuncture point group to build mould processing, in order to prevent thenatural recovery, normal group continues to conventional breeding.Adopt the method of ELISA and real-time fluorescent quantitative PCR observation spleendeficiency rats colon tissue of VIP, VIPR2, NO, NOS content and c-kit, SCF geneexpression levels.Results:1.Electric acupuncture "Zusanli","Sanyinjiao" VIP content in colon of rats.Results show that: compared with model group, after treatment with electricacupuncture, acupuncture point group rats colon VIP content is significantlylower than the model group (P<0.05),and close to the normal group, without goingthrough the acupuncture point group there was no evident difference with modelgroup, were significantly higher than in normal group.2. Cupping "Zusanli","Sanyinjiao" VIPR2gene expression levels of colon of rats.Results show that the amount of gene expression in model group1, comparedwith model group, normal group of colon VIPR2gene expression level of0.18±0.75times that of model group, acupuncture point set of colon VIPR2geneexpression quantity for the model group was0.27±0.90times, without goingthrough the acupuncture point group of colon VIPR2gene expression quantity for0.39±0.33times that of model group compared with model group, namely the colontissue VIPR2gene expression level in the acupuncture point group significantlylower than the model group, was statistically (P <0.05).3.Cupping "Zusanli","Sanyinjiao" colon of rats tissue changes in concentrationsof NO and NOS.Results show that: compared with model group, after treatment with electricacupuncture, acupuncture point group of colon tissue levels of NO and NOS weresignificantly increased(P<0.05),and close to the normal group, without goingthrough the acupuncture point group there was NO evident difference with modelgroup, were significantly lower than normal group.4.Cupping "Zusanli","Sanyinjiao" colon tissue of rat c-kit and the change ofSCF gene expression levels.Group compared with model group, the normal colonic c-kit,SCF gene expression level are respectively model group2.16±0.35and0.35±0.20times that ofacupuncture point set of model group was1.28±0.07and0.07±0.08times, withoutgoing through the cave group is a model of0.89±0.25and0.25±0.29times, namelythe colon tissue c-kit and SCF gene expression in the acupuncture point groupobviously higher than that of model group and without going through theacupuncture point group, with statistical significance (P <0.05).Conclusion:1.Electric acupuncture "Zusanli","Sanyinjiao" VIP content in colon of spleendeficiency rats and its receptor expression of VIPR2benign adjustment.2.Electric acupuncture "Zusanli","Sanyinjiao" to raise spleen deficiency ratscolon content of NO and NOS benign adjustment.3.Electric acupuncture "Zusanli","Sanyinjiao" of spleen deficiency rats colonSCF/c-kit signal way with improvement.4. Acupuncture "Zusanli","Sanyinjiao" of spleen deficiency rats colon barriershave improved effect.
