Objective:Taking the model rats with natural exposure and water conflictas the researching object, thebehaviors and the morphological changes in the related brain regions of Papez circuit will be observed, and the intervention effects of Chinese herb of Danzhixiaoyao decoction, which aims to help with the liver-discharging, heat-clearing and spleen-tonifying, will be explored.Methods:Evaluated by the open-field test, the 32 male Wistar rats were randomly divided into the normal group, the model group, the traditional Chinese medicine group and the western medicine group. Each group contains 8 rats. Rats with anxiety in the model group, the traditional Chinese medicine group and the western medicine group were formed by using natural exposure and water conflict. From the 15th day on, rats in the normal group and the model group began to be given a gavage ofdistilled water, and the rats in the western medicine group began to be given a gavage of buspirone hydrochloride, while the rats in the traditional Chinese medicine group began to be given a gavage of Chinese herb of Danzhixiaoyao Decoction. The continuous infusionsin four groups were implemented for 7 days. All the rats received the behavioral testing on the 7th,14th, and 21st day. Behind the last testing, samples of brains were acquired after 24 hours, and the related indexes were detected. Using the light microscope, the cell morphological changes in the neurons of the hippocampus, the amygdaloid nucleus, and the nucleus anterior thalami, which had been Hematoxylin-eosin stained, wereobserved. Using the electron microscope, the cell morphological changes of the neurons in the hippocampus and the amygdaloid nucleus were observed.Results:1. The behavioral results1.1 The open-field testCompared with the normal group, the crossing scores and the rearing scores of the model group, the traditional Chinese medicine group and the western medicine groupon the 7th,14th, 21st days all decreased. And, there exited significant differences (P< 0.05, P< 0.01).Compared with the model group, the crossing scores and the rearing scores of the traditional Chinese medicine group and the western medicine groupon the 7th,14th days were equal (P> 0.05).On the21st day, compared with the normal group, the score of model group had significant difference (P< 0.01), while the scores of the traditional Chinese medicine group and the western medicine group werethe same (P> 0.05); compared with the model group, the crossing scores and the rearing scores of the traditional Chinese medicine group and the western medicine groupboth increased, and the scores of the traditional Chinese medicine grouphad significant difference (P< 0.05).1.2Elevated plus maze testThe OE/TE (%) and OT/TT (%) values ofthe model group, the traditional Chinese medicine group and the western medicine groupon the 7th,14th,21st dayswere lower than those of the normal group, with significant difference (P< 0.05, P< 0.01). For the data of the 7th and 14th days,compared with the model group, the OE/TE (%) and OT/TT (%) values of the traditional Chinese medicine group and the western medicine group were the same (P> 0.05).For the data of the 21 st day,compared with the normal group, the data of the model group was significantly different (P< 0.05), while those of the traditional Chinese medicine group and the western medicine group were the same (P> 0.05);compared with the data ofthe model group, there were no differences except the OT/TT(%) value of the traditional Chinese medicine group (P<0.05)2. The observation results under light microscopeFor the rats of the normal group, the somas of the neurons of the hippocampus, the amygdaloid nucleus, and the nucleus anterior thalami were relatively big; the cell nucleus was quite big and round; the cytoplasm was pink and equally distributed. There were a large quantity of cells and they lined up in an orderly and concentrated manner. For the rats of the model group,the results showed the karyopyknosis of a large number of neurons, and they were hyperchromatic and deformed; the cytoplasm was condensed and dark red, and even some apoptotic bodies formed; the number of neuronsdecreased, and the cells were loosely arranged without rules. Such situation was particularly obvious in the CA2 and CA3 regions ofthe hippocampus. The cells of the traditional Chinese medicine group and the western medicine group had improved obviously compared with the model group, and their situation lied somewherebetween that of the normal group and that of the model group.3. The observation results under electron microscopeFor the rats of the normal group, the somas of the neurons of the hippocampus and the amygdaloid nucleus were relatively big; the cell nucleus was round or oval; the chromatin was equally distributed, and the organelles wereregular-arranged neatly. For the rats of the model group, the results showed the shrinking and deformation ofthe cell nucleus, and the double membrane structure of the nuclear membrane was unclear and even swelling and separation appeared; theintranuclearchromatin was loosely distributed; the heterochromatin increased and were flaky shaped and aggregatednear the nuclear membrane; the mitochondrion swelled or even ruptured, while the mitochondrial cristae disappeared; thecysts of the rough endoplasmic reticulum showed mild expansion. For the rats of the traditional Chinese medicine group and the western medicine group,there was obvious improvement and the shape of the cells lied somewhere in between.Conclusion:1.The activity and explorative behavior of the model rats with natural exposure and water conflict reduced. The encephalic region related to Papez circuit showed pathological injury. This model could simulate the GAD’s aetiological agent and statusbetter intrinsically. It will be beneficial to the medicine verification, and can become the platform for basic experimental research.2.The Chinese herb of Danzhixiaoyao Decoction, which is the main prescription for liver-discharging, heat-clearing and spleen-tonifying, can lessenthe anxiety behaviors of the model rats. Meanwhile, it can repair the encephalic region injury related to the rat’s Papez circuit and also facilitate the recovery. This point indicates thatthe Chinese herb of Danzhixiaoyao Decoctionhas obviousantianxiety effects.In these areas, it has similar curative effect with Buspirone Hydrochloride.
