Functional dyspepsia is a clinical common functional gastrointestinal disease, the clinical manifestations and to digest the bad as the main performance of the spleen deficiency syndrome is very similar, and in the clinical, functional digestion adverse, often in conjunction with spleen deficiency symptoms. Therefore, combination of disease and syndrome from angle to study the function of digestion adverse spleen deficiency syndrome has important significance. On functional dyspepsia spleen deficiency syndrome combination of disease and syndrome research rarely, and clinical studies are in the majority. Therefore, it is necessary to combination of disease and syndrome from Perspective of establ ishment and evaluation of functional dyspepsia spleen deficiency syndrome animal model at home and abroad. This paper will discuss the establishment and evaluation of the animal model of the deficiency of spleen deficiency syndrome by the literature review, theoretical research and experimental studies.Theoretical studyObjective:To establish and evaluate the animal model of the deficiency of spleen deficiency syndrome in the functional dyspepsia. Methods:Query system and function of dyspepsia and spleen deficiency syndrome related literature, brings forward the construction of functional dyspepsia spleen deficiency syndrome animal model, according to the experimental results, improve the making and evaluating methods for functional dyspepsia spleen deficiency syndrome animal model. Results:Functional dyspepsia literature search and method for making most of the literature problems, does not meet the functional dyspepsia Rome â…¢ diagnostic criteria and lack of evaluation method, in the literature at home and abroad, functional dyspepsia and TCM syndrome type associated, often and hepatic binding, the lack of functional dyspepsia spleen deficiency syndrome animal model. Spleen deficiency syndrome animal model establishment method and different, model evaluation is not uniform. Therefore, on the basis of the literature, functional dyspepsia spleen deficiency syndrome animal model establishment and evaluation principle: animal models must be both functional dyspepsia and spleen deficiency syndrome.Conclusion:Overstrain spleen deficiency syndrome is a common cause, iodoacetamide gavage cause increased sensitivity to visceral functional dyspepsia is important in the pathogenesis, so we are making use of iodoacetamide gavage functional dyspepsia disease, based on the use of Beijing small platform in the Department of diagnostic medicine University of Traditional Chinese Medicine Fashijialao tired standing over the TCM treatment of common factors, from the perspective of disease and syndrome successfully produced functional dyspepsia spleen deficiency syndrome in animal models.Experimental studyObjective:Production and evaluation of animal model of functional dyspepsia spleen deficiency syndrome. Method:The experiment was divided into two parts, namely:production and functional evaluation of dyspepsia spleen deficiency syndrome animal model, select quality model; the use of Xiangsha Liujunzi Decoction drug counterevidence. In the first experiment, we used healthy Spargue-Dawley male milk rats 180, and was born for 6 days. A total of 18 cages, each cage 10, with 1 rats, fed for 4 days. By the tenth day,180 rats were divided into control group (90 rats) and model group (90 rats).. The control group 90,2% sucrose solution by gavage,0.2ml/only; 90 rats in model group,0.1% iodoacetamide sucrose solution by gavage,0.2ml/only.6 days of continuous modeling, to the rats at 3 weeks of age, the sub cage,3-4 rats/cages. Normal feeding after 6 weeks of the model.. Rats at 6 weeks of age were were randomly divided into two groups, the control group were randomly divided into:normal group (n=45), small platform group (n=45), model group were randomized for:iodoacetamide group (n=45), iodoacetamide and small platform group (n=45). A small platform group and iodoacetamide plus small rats 17:00-7:00 platform every small platform stand, continuous modeling for 14 days. Daily use of function of digestive adverse spleen deficiency evaluation table recording and observing the rat hair, activities, body weight, food quantity, water intake, stool shape, symptoms and signs, record rats pull tail stool positive rate, detection obstacle dark experiment error frequency and latency, at the end of modeling after blood from the abdominal aorta, centrifuged to obtain serum were measured in rats serum amylase, lactic acid, d-xylose and motilin. In the second experiment, we use functional dyspepsia spleen deficiency syndrome type commonly used prescription:Xiangsha Liujunzi Decoction, drugs counterevidence, according to the experimental results of the first experiment, we selected iodoacetamide and small platform group as the optimization model and randomly assigned to 6 groups, respectively, for the high dose group of traditional Chinese medicine, Chinese medicine dose group, small dose of Chinese medicine group, western medicine group, model group and normal group. Large dose of Chinese medicine group (n=7): 0.2ml/kg Xiangsha Liujunzi Decoction irrigation stomach for 7 days; dose of traditional Chinese medicine group (n=7):0. lml/kg Xiangsha Liujunzi Decoction irrigation stomach for 7 days; traditional Chinese medicine small dose group (n=7):0.05ml/kg Xiangsha Liujunzi Decoction irrigation stomach for 7 days; western medicine group (n=7):domperidone irrigation stomach for 7 days, model group (n=7):lml/100g pure water irrigation stomach for 7 days; control group (n=10):lml/100g pure water irrigation stomach for 7 days. Daily use of function of digestive adverse spleen deficiency evaluation table records observed rat hair, activities, body weight, food quantity, water intake, stool shape, symptoms and signs, records of rat pull tail stool positive rate. At the end of modeling after blood from the abdominal aorta, centrifuged to obtain serum were measured in rats serum amylase, lactic acid, D-xylose, motilin. Results:In the first experiment, the table symptoms and signs were observed and the test of the indexes, in 14 days after modeling, iodoacetamide plus a small platform group rats appeared Shenpi fatigue, lethargy, lazy to move, hair loose and not Ze, weight increased slowly symptoms. Pull tail stool positive rate was significantly higher than that in the normal group, and normal group was statistically significant (P<0.05); serum amylase activity decreased, and the normal group compared to (P<0.05); lactic acid content increased, and the normal group compared to (P<0.05); serum D-xylose content decreased, and the normal group compared (P< 0.05). So we can conclude that iodoacetamide irrigation stomach and small platform is standing in line with functional dyspepsia spleen deficiency syndrome animal model in modeling requirements. In the second experiment, using Xiangsha Liujunzi Decoction treatment, traditional Chinese medicine high dose group, Chinese medicine dose group and small dose of Chinese medicine group rats symptoms have improved significantly, tend to normal stool, godless fatigue performance, hair is shiny and tidy, weight gain fast food intake increased rapidly. Large dose of Chinese medicine group and traditional Chinese medicine dose group of serum amylase, lactic acid content and motilin content close to the normal group, so it can be concluded that Xiangsha Liujunzi Decoction is effective in the treatment of. Conclusion:Iodoacetamide filling stomach with the small platform standing modeling method is consistent with functional dyspepsia spleen pathogeny, pathogenesis, course of disease, is qualified functional dyspepsia spleen deficiency syndrome animal model making method.
