Scleroderma sichuanensis Xiao and Scleroderma guani Xiao et Wu are both extern hymenopterous parasitoids (Hymenoptera: Bethylidae).,and they are the important natural enemy of the boring pests.This paper studied the cytology of the two species in order to provid necessary data for revealing the relationship,origin and evolvement between them;and it also analysed the genetic diversity between four types of differentiation' of Scleroderma sichuanensis Xiao and Scleroderma guani Xiao et Wu by using random amplified polymorphic DNA(RAPD) makers for the sake of studing the following srudies such as genetic breeding,which inchuding the period ,machanism,factors of variation and inducing good variation.Karyotype was analyed basing on the researching methods and chromosome technique in the thesis. The result were summarized as follows:1) The optimal production method:use the mature female ovaries of Scleroderma SPP as materials.The steps including: pretreated the materials in 0.01% colchicine liquo for 10 min,hypo-ingiltrated in distilled water for 5 min ,used Glacial acetic acid (3:1) fixed for 15 min ,and then dyed with Giemsa dye for 30 min , finally , exposed the flake in iced condition and then evelop it with neater balata.2) The chromosome number of the two species was 2n( ♀ )=30,and their karyotype formulas were 2n( ♀ )=4m+20sm+6st for Scleroderma sichuanensis Xiao and 2n (♀) =4m+18sm+8st for Scleroderma guani Xiao et Wu ,respectively.The karyotype of S.sichuanensis Xiao belonged to "3B" type of stebbins whereas Scleroderma guani Xiao et Wu belonged to " 2B " .Method of DNA extraction and reaction-system and optimized procedure of PCR Amplifition was reseached in the paper,and NTSYS2.1 software was used to analyse the data.The result were summarized as follows:1) The method on albumen enzyme K could get optimum DNA concentration, but fewer protein. It could be used to PCR Amplification.2) The optimal reaction of RAPD was studied , The optimum concentration of five important component such as Mg2+, Taq DNA polymerase, primer, template DNA, dNTP and DTT in 25ul RAPD reaction system were 3.5mmol/L, 1.5U, 0.lum/L,...
