Study On Technique System Of Tissue Culture In Daphne Odore Var. Marginnata

This paper mainly deals with the study on adventitious buds and Plantlets regenerated from sprout explants and leaves of Daphne odore Var.marginnata .The effects of medium component and cultivate approach on regeneration of Daphne odore Var:marginnata were studied by orthogonal and comparative experiment design.At the same time ,in the shoot approach, the effects of hormone and the others cultivated condition of effective multiplication, vitrification and chlorophyll accumulate were mainly studied .In the approach of callus, the hormone, part of explant,basis medium were mainly studied.The result showed that the optimum medium for various stages were as follows:(l)Initial medium: When combination of BA and IB A, MS+BA3 mg/L+IBA0.1 mg/L were the best medium .The shooting rate was up 68.86%.shoot high was up to 4.85cm. When combination of BA and NAA, MS+BA2mg/L+NAA0.1 mg/L were the best medium. The shooting rate was up 71.33%.shoot high was up to 5.32cm.The combination of BA and NAA was more beneficial for shooting than combination of BA and IBA.(2)Induction medium of callus: MS+BA0.5 mg/L+ KT2.0 mg/L+2, 4-D2mg/L+NAA2.0 mg/L+ sucrose3 % +agar0.8% were the best medium.(3)Polarization medium of callus : MS+BA2 mg/L +KT1 mg/L +2,4-D 0.2 mg/L +NAA0.4 mg/L were the best medium.(4) Proliferation medium: MS+NAAOmg/L or NAA0.1 mg/L+BA2 mg/L+ sucrose 40g/L+agarl.l% were the best medium. The effective multiplication ratio was up to 4 ~ 5,shoot was vigorous ,and contents of chlorophyll was higher under the optimum medium .The vigour of KT was inferior than B A, multiplication ratio of KT was lower than B A, but the shoot was more vigorous , the leaf extended ,the content of chlorophyll was high. AC restrained multiplication of test tube plant. Simultaneously in creasing cintent of agar ,the degree of vitrifiation of proliferation shoot can be decreased effectively, but the multiplication ratio was decreased.(5)Efficiency seedling induction medium: MS+BA0.5 mg/L +IBA0.1 mg/L +GA1.0 mg/L were the best medium.(6)Rooting medium: 1/2MS+IBA3.0 mg/L+sucrose2 % +active carbon0.2%were the best medium,in which rooting rate was 90%.(7)The survival rate of test-tube seeding was up to 65%by using vermiculite ,perlite and sand (1:1:2)as transplant medium.
