Study On Selection Of Vaginal Microbes Therpying Dairy Chronic Endometritis

In this research, it includes six apartments of test: isolating and identifying of microbes in normal vaginal ; segregating and identifying of bacterial pathogen ; antagonism test in vitro;test-ing of immunogenitity of isolated strain ;safety test ; experiment of establishing pathological model and therapeutic action .1) Samples are collected from three multiparious dairy vaginal, which are used to isolate and purificate different microorganisms. We determine several sorts of bacteria such as staphylococcus.epdermidis; Citrobacter; Lactbacillusacidophius; Neisseria.sicca; Escherichia. coli;Leucono-stocmes. entaroides;Brevibacillu. spolymyxa; Brevibacillus. brevis; Streptococcus.agalactiae; Staphylo-coccus.aureus.Peptostreptococcus by series of morphological dentification and biochemical tests .2) Selective mediums are available to screen three strains of pathogenic microbial such as Streptococcus;Escherichia.coli;Staphylococcus.aureus whose pathogenic action on body is not strong.The strain of Streptococcus presents a hemolysis on streaking blood plate .In coagulase-positive Staphylococcus.aureus group ,the result that both survive is same as that mEscherichia.coli group.3) The filter and culture fluid have an antagonistic action on Escherichia. coli.Strept coccus; Staphylococcus.aureus; in different level. The sequence of intension of filter fluid is B1, A1, C6,C1 ,C5,B3 ,A3,C2,B5,C3,B2 ,A2 while that of culture fluid is B1 ,A1,C6,A3, C5 ,C1, C3, B2 , C2,B5 ,A2,B3 -There has a different significance (P< 0.05) in antagonism test on three kinds of bacteria between B1 and some other strains .According to judging standard of antibiotic inhibition ring, B1 A1 are highly susceptibility to pathogen of chronic endometritis ,some other are moderate excluding B2, C2, A2.4)Four strains of isolated bacteria can enhance immune function of rabbit. After vaccinating isolated bacteria culture,serum level of IgG, IgA, IgM presents ascending trend .There is a significant difference in serum level of A1 strain compared with other three strains and control group after vaccinating.while IgA serum level of B1 has higher significant difference than someother strains5) It proves that four isolated strains of A1, B1, B5, C5 are safe on body through safety test, The result in acute and subacute toxicity test of four strains shows that there is not bad effecton ten mice fed in different concentration of culture fluid after firstly per os and successive drinking for thirty days . Finally ,breeding of mice is normal and pathologic lesion of them is found after anatomy check.Judging by food safety standard in toxicology, toxical action of four isolated strains of microbial on mice is evaluated as nonvirulent dosage grade in LD50=501~5000mg/kg.6) Rabbits are cured of vagintis in pathologic model,which are injected into vigina with culture fluid of Lactbacillusacidophius and mixture of penicillin and streptomycin differently for three time.Result shows that 70% and 87.5% of cure rate between them.
