It can hydrolysis of extracellular matrix in a variety of ingredients.Thefunction of MMP-2 is to organize and restore ECM. TIMP – 2,anatural tissue inhibitor of MMP – 2,can combine both active and inactive MMP – 2 to depress the activities of MMP- 2, so that the activities of MMP- 2 suspended, after it is very useful to make the activity of collagen and gelatin hydrolysate of MMP- 2 reduced, eventually make the movement and attack cancer cells or inhibit endometrial cells. Test according to the survey, in normal circumstances, in the interaction of MMP- 2 and TIMP- 2, coordinate with each other. Interleukin 10 is a Th2 secreted by macromolecular; can reduce the function of Th1;and has a strong anti-inflammatory effect; it isan anti-inflammatory factor. Prostaglandin E2(PGE2) is a kind of proinflammatory cytokines, it can not only promote the Th1 cells and Th1 type of T cell differentiation, then promote inflammation; but also can inhibit the immune response and cell to gobble up,neutrophil accumulation and migration. Research shows that IL – 10,PGE2 and mammals in MMP – 2,TIMP- 2 are closely linked, cow endometrial the degradation of ECM and restructuring of MMP- 2 and TIMP- 2 close control. Due to the MMP- 2 and TIMP- 2 effects of cow endometritis, can control the occurrence of cow endometritis development and disease prevention.Theexperiment provides a new theoretical basis and experimental basis.This experimentchooses the healthy Holstein cows for a control group and experimental group for endometritis Holstein cows each 10 head.I take their blood and endometrial tissue. For interleukin 10 and prostaglandin E2 concentration in serum was determined by ELISA kit. Part of organization from which is used to extract total RNA.They after the reverse transcription use fluorescence quantitative PCR method to detect matrix metalloproteinases-2 and matrix metalloproteinases inhibitor-2 m RNA expression level. The other part is used in the immunohistochemical determination of matrix metalloproteinases 2 and matrix metalloproteinases tissue inhibitors- 2 at the level of protein secretion. The data is to use SPSS Statistics 17.0 for the analysis of experimental results.The experimental results show that:1. Concentrations of PGE2 in healthy Holstein cows group was1.47 3 103±80.29ng/L; IL-10 content was48.41±5.45 ng/L. The group of Holstein cows with endometritis concentrations of PGE2 was0.933103±21.99 ng/L; IL-10 content was78.45±2.55 ng/L. The concentration of PGE2 in the experimental group was significantly lower than the control group(P < 0.01); but the content of IL- 10 is significantly higher than that of control group(P < 0.01).2. Matrix metalloproteinases-2and tissue inhibitor of metalloproteinase-2 of m RNA levels in experimental group endometrial tissue were significantly lower than the control group(P<0.01).3. Matrix metalloproteinases-2 and tissue inhibitor of metalloproteinase-2 in healthy group of protein levels expressed in IOD values, and they respectively were(10.452±1.231)3103ã€(8.367±1.324)3103;While suffering from endometritis group respectively were(5.432±0.587)3103ã€(4.287±0.536)3103. Thus, MMP- 2 and TIMP- 2 in healthy group of protein expression levels were higher than the control group.The difference was significant(P < 0.01).Experimental results show that in the endometritis the content of IL- 10 cattle raised, reducing the content of PGE2.They caused the endometrium of MMP- 2 and TIMP- 2 lower expressions; causing the ECM supply cow balanced steady state of endometrial cells get damaged, and promoted the emergence and development of endometritis.
