Study On Technique System Of Tissue Culture In Cinnamomum Septentrionale Hand.-Mzt

The efficient method was explored about Cinnamomum Septentrionale Hand.-Mzt in Lauraceae in vitro Multiplication. The stems with bud of Cinnamomum Septentrionale Hand.-Mzt plantlet and mature tree were used for to establish the technich system of tissue culture. Multydesign and multyanalysis was used to find out the best explant x proceeding of culturing and the best composition of medium.The results were as follows:(1)Selecting explant: bottom axillarry buds as explant is effect to childhood . Butterminal buds is effect to mature age(2)Iniation culture:MS medium with BA2.0mg/l,NAA0.01mg/l and sugar 30g/l,is the best iniation medium to sprout to childhood.(3)Subcultuie culture:the MS medium with the BA6.0mg/l and NAAO.Olmg/1 was applied as the subculturing medium,and the increasing coefficient in one month is 5.9, using 30 days as subculture time,theoretically,the times of propagation is 109. (4)Using 1/2 MS with N A AO. 01 ml/1, stronger nurslings could be got. (5)Rooting culture: 1/2MS mediumwithIBA2.0mg/L,NAA0.1mg/l is the best one in rooting culture.the rooting rate is 86.00%.(6)Transplantion:the transplanting medium in perlite and vermiculite bylto l,is useful.
