| Cymbidium grandiflorium is a kind of highly ornamental Orchidaceae plant. It was propagated by means of offshoots trationally. With the development of modern biological technology, tissue culture could be gradually applied to it's propagation, which could greatly increased it's propagation coefficient. However, the program and medium of micropropagation should be simplified in order to reduce the cost in mass production. This paper compared the effects of inducement of protocorm-like bodies (PLB) in the different medium with stem apex as explants, and of multiplication and differentiation of PLB in different medium with PLB as explants. The results were as follows:(1) The best medium for the inducement and multiplication of PLB both were 1/2MS+BA 1.0mg/L+NAA 0.5mg/L, so it could reduce a medium preparation; modified KC+KT 0.05mg/L+NAA 0.5mg/L was also could be used. They could be chosen to use in practice.(2) The best medium for the developing of shoots from PLB was 1/2MS+BA 1.5mg/L+NAA0.5mg/L.(3) The best medium for the developing of roots from PLB was 1/2MS+NAA 0.1mg/L+GA1.0mg/L.(4) It was workable that white sugar replaced sucrose and tap water replaced distilled water. Simplified medium further reduced production cost.At the same time, to apply Cymbidium grandiflorium to decorate and beautyindoors better, another part of this paper was with an eye to it's soilless cultivation techniques. The results were as follows:(5) The best transplant medium of Cymbidium grandiflorium seedlings was haydites with barks or pseudopodium; the best nutrient fluid was Ca(NO3)2 472m/L+KO3 809mg/L+KH2PO4 153mg/L+MgSO4 493mg/L. |
