Abstract/Summary:
Flos Pueraria omeiensis is one of the earliest medicinal plants used in traditional Yao' s folk-medicine . Flos Pueraria omeiensis & Ilex latifolia(PL) decoction is often used for antialcoholism and treatment of hepatitis. Its chemical components and their pharmacological effect and function mechanism have never been reported. Thus the extraction, separation and structural identification of Flos Pueraria omeiensis were carried out. And by establishing in vitro antibacterial and animal model, we studied the correlations between the chemical components and their bioactivities, and the mechanism of antialcoholism and protective action on the liver. These results provide the scientific basis for the development and application of Flos Pueraria omeiensis and PL decoction.Five compounds were isolated from the ethyl acetate fraction of ethanol extract of Flos Pueraria omeiensis. And Four steroids were identified by GC/MS technique. They were tectorigenin, luteolin, genistein , dimethyl tectorigenin , daidzein , Cholest-5-en-3 β -ol , Ergost-5-en-3-ol, stigmasterol, sitosterol. One compound was determined as tectoridin from the n-butanol section. These compounds are found in Flos Pueraria omeiensis, and luteolin is found in the plants of Puerarea DC. for the first time.The content of Ca, Mg, Cu, Zn, Fe and Mn was determined in Flos Pueraria omeiensis and its decoction by Flame Atomic AbsorptionSpectrophotometry. The results showed the content of six inorganic elements of Flos Pueraria omeiensis was similar to that of the roots of P. lobata. The essential oil of Flos Pueraria omeiensis was extracted by steam distillation. Fifty-nine chemical constituents in the essential oil were identified by GC-MS, and the main components are terpene.To study the antibacterial effect of four crude extracts, some sections isolated from ethyl acetate fraction ,the essential oil and three compounds from Flos Pueraria omeiensis by MIC method. Tectoridin and essential oil did not inhibit bacterial activity. However, tectorigenin and genistein inhibited bacterial activity.In the rat model of acute alcohol and CCU hepatic injury, we studied the function of antialcoholism and protective action on the liver of Flos Pueraria omeiensis and PL decoction in the preliminary experiments. The result shows, drinking Flos Pueraria omeiensis and PL decoction before drinking liquor can prevent alcoholism, taking them after drinking liquor can relieve alcoholism. They are more effective in preventing alcoholism than in relieving alcoholism. Flos Pueraria omeiensis and PL decoction were more effective than Haiwang Jinzhun in the preliminary experiments. The low and high doses of Flos Pueraria omeiensis and high dose of PL decoction all had protective effect on the liver.
