Puerariae Radix (P.R), a kind of both medicinal and edible plant which has been approved by the State Ministry of Health, is widely cultivated and utilized in China. In the paper, PR slices are taking as the raw material to do the following researches:The ingredients of P.R were measured as the national standard method. The results showed that there were a total isoflavone of7.45mg/g, a puerarin of3.37mg/g, a starch of412.30mg/g, a protein of46.23mg/g, an ash of20.25mg/g, and a adipose of6.33mg/g in P.R.The effect on P.R total isoflavones yield of five single factors including ethanol concentration, solid-liquid ratio, temperature, time and times of extraction was studied by single factor experiments. Then using the total isoflavones as evaluation index to determine the optimal extract parameters by orthogonal experiment. The optimal process were: temperature75℃, solid-liquid ratio11Bv, ethanol concentration60%(V/V), extraction time2.5h, extraction times twice. Under this optimal condition, the content of total isoflavones in crude extracts was37.9%and the average extraction rate was71.5%. The puerarin content in crude extracts was15.8%.The total isoflavones were separated from crude extracts by AB-8macroporous resin column. It got the total isoflavones preparation that had88.2%total isoflavones, which had38.2%puerarin. The puerarin of98.2%purity was separated from crude extract by silica gel column chromatography.The results of total isoflavones and puerarin of P.R on acute hangover efficacy in mice showed that both of them could significantly prolong the ebriety time and shorten the sober up time of mices. What’t more, the effect of total isoflavones was better than the same dose puerarin.In the experiment of total isoflavones on mice chronic alcoholic liver disease preventive, the mices which were long-term fed different doses of drugs and alcohol were observed in liver biopsy. And multiple indicators of blood and liver tissue were also detected.The results showed that the total isoflavones can increase the liver ADH, GSH enzyme activity, decrease MDA content in liver, accelerate the metabolism of alcohol in the liveral, reduce the generation of toxic metabolic intermediates, reduce the risk of alcohol-induced liver injury. The indexes of ADH, ALDH, GST, and MDA in blood were significantly ruduced, and the GSH-Px and SOD indexes were increased. It indicated that total isoflavones can effectively enhance the capacity of liver to metabolize of alcohol, reduce the amount of unmetabolized alcohol into the blood circulatory system. It played a role in protecting the body.The optimize P.R beverage recipes of sober-up and liver-protection function was abtained by single factor and orthogonal experiment, which were total isoflavones2%, rock sugar7%, and citric acid0.1%. The production process and product related sensory indicators, physical and chemical indicators and health indicators were developed. And all the indicators were detected. The result of acute toxicity test was LD5o>600mL/(kg·mb·md). Functional verification showed that the ebriety time was6.73mins longer and the sober up time was84.25mins shorter. And hence, the sober-up and liver-protection functional beverage of Pueraria could protect liver from alcohol and drunkness.
