The technology of extracting total saponins from the bulbs of lilium brownii was studied .The orthogonal design was used to optimize the extraction process of lilium brownii with content of total saponins. The result showed that the optimum extraction conditions obtained were temperature 70 C, 80% alcohol, solid/liquid of 1 : 6 and reflux time of 3 h with 3 times. The order of these five factors on affecting the extracting yield of total saponins was: E>A>D>C>B. In the condition,the concentration of total saponins was 3.48%.An accurate method for determination of the content of total saponins in lilium brownii was reported. The dioscin was used as the control sample, the vanillin-perchloric acid as chromogenic agent, glacial acetic acid as solvent system at 60Cfor 15min.Then,the content of total saponins in lilium brownii were determined by spectrophotometry at 540nm.The calibration curve had good linearity, r=0.9996. The recovery ratio of measurement was 96.74%~ 103.65%, and the relative standard deviation was 3.0%. The method had good sensitivity and reappearance.Three kinds of macro porous adsorbents, "X-5, AB-8 and NAK-9 were studied on adsorption performance for total saponins of lilium. It was showed that AB-8 was a good adsorption resin for total saponins of lilium. Technological process of purification was studied with the macro porous adsorbent of AB-8. The optimum conditions were: after extracting coarse solution of saponins, adsorption and separation by AB-8 resin with 70% ethanol, which was used as eluted solvent.The study was performed to investigate the radical scavenging activity of extracting of lilium. Bromopyrogallol red(BPR) was used as chromogenic reagent. A system Co2+~H2O2 produces hydroxyl radical ( OH) by reaction between Co2+ and H2O2 while the hydroxyl radical change the color of BPR. The amount of -OH can be indirectly assayed through absorbance change by means of UV-Vis spectrophotometer. The Hydroxyl radical scavenge activity of extracting of lilium was evaluated. The results showed the purify extract of lilium had higher ability toscavenge hydro-radical than that of the coarse extract of lilium.
