Bioleaching is a kind of technology, especially suitable for low grade ores, overburden and waste from current mining operation, by using microorganism to recovery metal from mineral. Bioleaching is generally more environmentally friendly than conventional metal recovery processes such as concentration. Comparing to bioleaching by one kind bacteria, using a mixed culture is more and more efficiency. To construct a more effective mixing leaching, it is of great significance to understand the change of community system between the process of ore mineral extraction. The aim of the study is to find a kind of method to judge the process of leaching in mixed community system. For Real-time PCR in the microbial community system, the paper analyzes the change and effect is studied.For the mixture of the eight microbial culture, the eight pairs of specific primers are synthesized and evaluated. According to the 16s RNA, gyrB, soxB or arsB gene of different bacteria these primers are designed. Through the comparison analysis of BLAST sequence, the results of PCR product fragment size showed that the Ct values and melting curves demonstrated a single peak both primer specificity and sensitivity.For the same samples separately with Real-time PCR and RFLP two methods to analysis both for Leptospirillum ferriphilum is dominated strains. Relative to RFLP, Real-time PCR has some advantages, for example more short detection time, detection sensitivity and other.In pH gradient leaching test, obtaining from the process to the DNA samples are dectected by Real-time PCR. The first 24 days, Acidithiobacillus caldus is a advantage of both strains. In contrast, Acidithiobacillus ferrooxidans has been declining, while the proportion of t Leptospirillum ferriphilum is increase in their total amount of bacteria. This phenomenon may be due to the reduction of Fe2+ or the change of Fe3+ concentration.
